The objective of the present study is to investigate the animal risk factors related to the presence of E. coli O157:H7 in feces and milk samples collected from healthy cattle at differing ages in private and government dairy farms in Kayseri and its vicinity. Five hundred fecal samples and 500 raw milk samples were collected from different locations in Kayseri, processed through immune Magnetic Separation, and inoculated Chorom medium and Sorbitol MacConkey medium. Suspicious colonies were selected from this medium, and they were examined serologically with H and O agglutination test. The molecular direction suspicious isolates were examined using multiplex with PCR (mPCR) and uniplex PCR (uPCR) techniques. The examination showed that only 1 (2 %) out of 500 raw milk samples and only 6 (4.6%) out of 500 fecal samples contained O157:NM strain, and none of the samples contained flagella H antigen. Through using mPCR, only one sample from O157:NM of isolated fecal samples contained Ehly A, whereas only one sample contained stx2 genes. None of the other fecal samples contained any virulence genes. Only one raw milk sample isolated from O157:NM strain did not have any virulence genes.In conclusion, the raw milk and fecal samples collected from dairy farms in Kayseri region and its vicinity contain E. coli O157. Low levels of contamination rate was determined in the samples. However, it was concluded that due attention should be paid to hygiene rules in Kayseri
