The purpose of this study was to investigate the effect of laboratory activities designed based on Predict-Observe-Explain (POE) strategy on pre-service teachers' understanding of "Substance Transport in Plants" in the Science and Technology Laboratory-I course. In this study, the sample consisted of 144 pre-service primary teachers who took the laboratory course Faculty of Education at Pamukkale University. The research was realized in fall semester of 2010-2011 academic year. Results of the analyses showed that the effect of activities based on POE strategy on pre-service primary teachers' conceptual achievements. At the end of the study it was shown that misconceptions about the concept of transporting in plants the POE strategy was effective in identifying students` misconceptions and teaching the concept. It was suggested that POE strategy could be applied in other topics in science and also in other disciplines. But their effectiveness should be investigated.
