Bu çalışma, streptozotosin (STZ) ile indüklenen diabetik ratların, böbreklerinde oluşan histolojik değişiklikler üzerine ginkgo bilobanın etkilerinin araştırılması amacıyla planlandı. Çalışmada Wistar Albino cinsi 40 adet erişkin erkek rat kullanıldı. Ratlar, kontrol, diabetik grup, Ginkgo verilen diabetik grup ve çözücü verilen grup olmak üzere 4 gruba ayrıldı. Deneysel Diabet, diabetik ve Ginkgo biloba verilen diabetik gruplarda tek doz STZ'nin (60 mg/kg) intraperitoneal uygulanmasıyla oluşturuldu. Diabet oluştuktan sonra, Ginkgo biloba grubuna 40 gün süresince her gün 100 mg/kg ginkgo biloba ekstresi oral olarak verildi. Deney süresince ratların kan-glikoz seviyeleri ve vücut ağırlıkları takip edildi. 40. gün ratlar eter anestezisi altında dekapite edilerek böbrekler alındı. Dokular rutin takip sonrası, parafine gömüldü. Histokimyasal boyama sonrası, kesitler ışık mikroskopta incelendi. Diabet grubundaki ve Ginkgo verilen gruptaki ratların, kontrol ve çözücü verilen gruba göre kan- glikoz seviyeleri önemli oranda artarken, vücut ağırlıklarıda önemli oranda azaldı. Diabetik grupta glomerüllerde mezengial matriks genişlemesi bazal membran kalınlaşması, glomerüloskleroz, tübüllerde vakuolizasyon, tübüler nekroz, hidropik dejenerasyon ve hücre şişmesi gibi temel histolojik değişiklikler gözlendi. Uygulanan Ginkgo biloba ekstresi ile bu bulguların önemli derecede azaldığı tespit edildi. Ginkgo biloba ekstresi (EGb 761), Streptozotosinle ratlarda oluşturulan diabetin sebep olduğu böbrek hasarlarını azalttı. Bu yüzden Ginkgo biloba ekstresinin diabetik böbrekte görülen patolojik bulguları azaltacağını ya da gelişimini önleyebileceğini düşünmekteyiz. Yinede diabetik komplikasyonlar üzerindeki yararlı etkisini gösterebilmek için uzun süre kullanımıyla ilgili, ileri düzeyde çalışmaların yapılması gerekmektedir.
This study was planned to investigate the effects of ginkgo biloba on histological changes in the kidneys of diabetic rats triggered by streptozotocin (STZ). In the study, 40 adult male rates of the Wistar Albino genre were used. It was divided into 4 groups, including rats, control, diabetes group, Ginkgo given diabetes group and solvent given group. The experimental Diabetes, Diabetes and Ginkgo biloba diabetic groups were created with single dose of STZ (60 mg/kg) intraperitoneal application. After diabetes, the Ginkgo biloba group was given 100 mg/kg of Ginkgo biloba extract per day for 40 days. During the test, the blood glucose levels and body weight of rats were monitored. On the 40th day, rats were decaptivated under ethereal anesthesia and kidneys were taken. After a routine follow-up, the tissue was buried in the paraffin. After histochemical painting, the cuts were studied in the light microscope. Rats in the diabetes group and Ginkgo given group, blood glucose levels increased significantly according to the control and solvent given group, while body weight decreased significantly. In the diabetic group, basic histological changes such as the thickness of the basal membrane, glomerulosclerosis, vacualization in tubules, tubular necrosis, hydropic degeneration and cell swelling were observed. With the Ginkgo biloba extract applied, these findings were found to be significantly reduced. Ginkgo biloba extract (EGb 761), reduced kidney damage caused by diabetes created in rats with streptozotocin. That is why we believe that Ginkgo biloba extract will reduce or prevent the pathological symptoms found in the diabetic kidney. Instead, advanced studies on long-term use are needed to show its beneficial effect on diabetic complications.
