GİRİŞ ve AMAÇ: Bu çalışmanın amacı, osteoporotik sıçanlarda kortikal kemik defektlerinde hyaluronik asit ve dentin greftinin kemik iyileşmesi üzerindeki etkisini değerlendirmektir. YÖNTEM ve GEREÇLER: Çalışmada 24 adet erişkin dişi sıçan kullanılmıştır. Sıçanlara bilateral ovarektomi yapılmış, altı hafta sonunda osteoporotik olarak kabul edilen sıçanlar, üç gruba ayrılmıştır. Grup D, Grup D-HA ve Grup C. Her hayvanın kalvarium’unda 5 mm çapında kritik boyutta bir defekt oluşturulmuştur. Defektler D grubunda; dentin grefti ile, D-HA grubunda; dentin grefti ve hyaluronik asit ile doldurulmuş, kontrol grubunda (C grubu) ise boş bırakılmıştır. Bütün hayvanlar 28. günde sakrifiye edilmiştir. Her defekt bölgesinde yeni kemik oluşumu; hematoksilen ve eozin, BMP2, TGFβ ve osteopontin seviyelerine göre hem histolojik hem de immünohistokimyasal olarak değerlendirilmiştir. BULGULAR: Histolojik olarak grup D ve grup D-HA, grup C’den anlamlı olarak daha fazla yeni kemik oluşumu göstermiştir. İmmünhistokimyasal olarak; TGFβ ve osteopontin seviyeleri Grup D-HA’da anlamlı olarak yüksek bulunmuştur. BMP2 seviyeleri D grubu ve D-HA grubu arasında anlamlı farklılık göstermemiştir. TARTIŞMA ve SONUÇ: Dentin grefti ve hyaluronik asitle birlikte dentin greftinin osteoporotik kemiklerde yeni kemik oluşumunu hızlandırdığı gözlenmiştir.
The aim of this study is to evaluate the effect of hyaluronic acid and dentin in cortical bone defects in osteoporotic rats on bone healing. Method and Requirements: 24 adult female rats were used in the study. The rats, which have been carried out bilateral ovarectomy, considered osteoporosis at the end of six weeks, are divided into three groups. Group D, Group D-HA and Group C. A critical size defect is created in the calvarium of each animal with a diameter of 5 mm. The defects are in group D; with the D-HA group, with the D-HA group; filled with the D-HA group and hyaluronic acid, and left empty in control group (Group C). All the animals were sacrificed on the 28th day. The new bone formation in each defect area was assessed both histological and immunohystochemical according to the levels of hematoxylene and eosine, BMP2, TGFβ and osteopontine. Histologically, group D and group D-HA showed significantly more new bone formation than group C. Immunochemically, the levels of TGFβ and osteopontin were significantly high in Group D-HA. The BMP2 levels have not shown any significant differences between group D and group D-HA. DENTIN INGREPT AND HYALURONIC ACID: The DENTIN INGREPT, along with DENTIN INGREPT and HYALURONIC ACID, has been observed to accelerate the formation of new bones in the osteoporosis bones.
