Allium cepa L.’ da Krom (K2Cr2O7)’un Toksik Etkilerinin Araştırılması

Bu çalışmada, farklı dozlarda Krom ağır metalinin (K2Cr2O7) Allium cepa L.’de fizyolojik, sitogenetik ve anatomik etkileri araştırılmıştır. Fizyolojik parametre olarak çimlenme yüzdesi, kök uzunluğu ve ağırlık artışı; sitogenetik parametre olarak mikronukleus (MN) sıklığı, kromozomal aberasyonlar ve mitotik indeks (MI) araştırılmıştır.  K2Cr2O7 uygulaması ile çimlenme yüzdesi, kök uzunluğu, ağırlık artışı ve MI oranlarının azaldığı, MN sıklığı ve kromozomal hasarların ise arttığı belirlenmiştir. En belirgin toksik etki ise 12.5 mg/L K2Cr2O7 uygulanan grupta gözlenmiştir. Mikroskobik incelemeler neticesinde K2Cr2O7’nin teşvik ettiği kromozomal hasarlar; kromatinin eşit olmayan dağılımı, fragment, yapışkan kromozom, kromozom köprüsü, c-mitoz, ters kutuplaşma olarak belirlenmiştir. K2Cr2O7 uygulamasının kromozomal hasarlar içerisinde en büyük etkinin fragment oluşumu üzerine olduğu tespit edilirken, en az oranda gözlenen kromozom hasarının ise ters kutuplaşma olduğu belirlenmiştir. Ayrıca K2Cr2O7 uygulaması kök ucu meristematik hücrelerinde yassılaşmış hücre çekirdeği, korteks hücre çeperinde kalınlaşma, hücre deformasyonu ve nekroz şeklinde anatomik hasarlara neden olmuştur. Sonuç olarak, K2Cr2O7’nin A. cepa’da fizyolojik, sitogenetik ve anatomik açıdan ciddi toksik etkilere neden olduğu, bu etkinin de doza bağımlı olarak gerçekleştiği belirlenmiştir.

In this study, the physiological, cytogenetic and anatomical effects of chromium heavy metal (K2Cr2O7) in different doses were studied. As a physiological parameter, the percentage of grinding, root length and weight increase; micronucleus (MN) frequency as a cytogenetic parameter, chromosomal aberrations and mitotic index (MI) have been studied.  The application of K2Cr2O7 determined that the percentage of grinding, root length, weight increase and MI rates decreased, and the frequency of MN and chromosomal damage increased. The most noticeable toxic effect was observed in the 12.5 mg/L K2Cr2O7 group. As a result of microscopic examinations, the chromosomal damage promoted by K2Cr2O7 was determined as the uneven distribution of chromatine, fragment, adhesive chromosome, chromosome bridge, c-mitosis, reverse polarization. The application of K2Cr2O7 has been found to have the greatest effect on fragment formation in chromosomal damage, while the least observed chromosomal damage has been found to have the reverse polarization. Also, the application of K2Cr2O7 has caused anatomical damage in the form of a flat cell core in the root end meristematic cells, thickness in the cortex cell circle, cell deformation and necrosis. As a result, K2Cr2O7 has been determined to cause serious toxic effects in a physiological, cytogenetic and anatomical aspect of the A. cephe, and this effect occurs depending on the dose.
