Bu çalışmada, su kalitesini analiz etmek için Allium testi kullanıldı. Civil deresinden alınan yüzey suyu örneklerinin potansiyel toksik etkileri Allium cepa L. tohumlarında çimlenme yüzdesi, kök uzunluğu, tohum ağırlığı, mitotik indeks (MI), kromozomal anormallikler, mikronukleus (MN) sıklığı ve anatomik değişimler gibi toksisite indikatörleri tarafından araştırıldı. Tohumlar kontrol ve dere suyu uygulama grubu olarak iki (2) gruba ayrıldı. Kontrol grubundaki tohumlar çeşme suyu ile muamele edildi. A. cepa L. kök ucu hücreleri Feulgen reaksiyonu ile hazırlandı ve Asetokarmin ile boyandı. Ağır metal analizleri için su örnekleri toplandı ve İndüktif olarak Eşleştirilmiş Plazma-Kütle Spektrometresi (ICP-MS) kullanılarak analiz edildi. Sonuçlar, kontrol grubu ile karşılaştırıldığında, dere suyu ile muamele edilen A. cepa L. tohumlarının çimlenme yüzdesi, kök uzunluğu, tohum ağırlığı, mitotik indeks (MI), kromozomal anormallikler ve mikronukleus (MN) sıklığında istatistiksel açıdan önemli (p<0.05) değişmeler olduğunu gösterdi. Ayrıca, dere suyu A. cepa L. kök uçlarının hücresel yapılarında ciddi hasarlara sebep oldu. Dere suyunda vanadyum (V), krom (Cr), kobalt (Co), nikel (Ni), bakır (Cu), çinko (Zn), arsenik (As), kadmiyum (Cd), titanyum (TI), kurşun (Pb) ve demir (Fe) gibi ağır metaller belirlendi. Sonuçlar, dere suyundaki kirliliğin çevre ve canlı organizmaların sağlığı için bir tehlike ve ikaz olarak düşünülmesi gerektiğini gösterdi.
In this study, the Allium test was used to analyze the water quality. The potential toxic effects of surface water samples taken from the Civil Seed were studied by toxicity indicators such as grinding percentage, root length, seed weight, mitotic index (MI), chromosomal abnormalities, micronucleus (MN) frequency and anatomical changes in the seeds of Allium seed L. The seeds were divided into two (2) groups as a control and drilling water application group. The seeds in the control group were treated with spring water. A. cepa L. root end cells were prepared with the Feulgen reaction and painted with acetocarmin. Water samples were collected for heavy metals analysis and analyzed using Inductively Combined Plasma-Mass Spectrometer (ICP-MS). The results, compared to the control group, showed that the seeds of A. cepa L. treated with drunk water had statistically significant variations (p<0.05) in grinding percentage, root length, seed weight, mitotic index (MI), chromosomal abnormalities and micronucleus (MN) frequency. Furthermore, drill water caused serious damage to the cell structures of the A. cepa L. root ends. In Dere water, heavy metals such as vanadium (V), chromium (Cr), kobalt (Co), nickel (Ni), bakery (Cu), zinc (Zn), arsenic (As), cadmium (Cd), titanium (TI), bullet (Pb) and iron (Fe) were identified. The findings showed that pollution in water should be considered as a danger and indication for the health of the environment and living organisms.
