Kuzeydoğu Anadolu ve Kuzeydoğu Karadeniz Bölgelerinde Yayılış Gösteren Kafkas Arı Irkı (Apis mellifera caucasica) Hibritleşme Düzeyinin Saptanması

Bu çalışma Dünyada ki arı ırkları üzerinde yapılan çalışmalar sonucunda Türkiye’de ki doğal lokasyonlarının Kuzey Doğu Anadolu ve Doğu Karadeniz bölgeleri olduğu bildirilen Kafkas bal arısı (A. mellifera caucasica) ‘nın hibritleşme düzeyinin PCR tekniği ve filogenetik ağaçlandırma metodu kullanılarak tespit edilmesi amaçlanmıştır. Popülasyonlar içerisinde ki seleksiyonların türler üzerinde yarattığı genotipik etkinin düzeyini ölçmek için mitokondriyel DNA (mtDNA) molekülünün Sitokrom C Oksidaz I geni baz alınmıştır. Kuzey Doğu Anadolu ve Doğu Karadeniz bölgelerinde bulunan Kars, Ardahan, Erzurum Artvin ve Rize illerinin popülasyonları hedef olarak seçilmiştir. Çalışma materyali olarak bu illerin sınırları içerisinde bulunan 57 arılıktan yaklaşık 450 ergin işçi arı örneği (her istasyondan yaklaşık olarak 7-8 adet arı örneği alınmıştır) toplanmıştır. Arılıklardan toplanan ergin işçi arı örneklerinin kalıtsal materyalleri fenolklorofrom: izoamilalkol tekniği kullanılarak izole edilmiştir. İzolasyonları yapılan DNA örneklerinin PCR sonrası elde edilen ürünleri jel elektroforezinde yürütüldükten sonra agaroz jel görüntüleri alınarak dizi analizleri yapılmıştır. Dizi analizi verileri sonucu materyallerin akrabalık düzeyleri filogenetik ağaçlandırma modellinde değerlendirilmiştir.

Abstract:

This study is aimed at identifying the level of hybridization of the Kafkas honey (A. mellifera caucasica) that its natural locations in Turkey are north-eastern Anadolu and eastern Black Sea regions using the PCR technique and the philogenetic method of treening. In the populations that selections create on species genotype effect levels are measured by the cytokrom C oxidaz I gene of the mitochondrial DNA molecule (mtDNA). The populations of the provinces of Kars, Ardahan, Erzurum Artvin and Rize, located in the northeastern Anadolu and eastern Black Sea regions, have been selected as a target. As the work material, approximately 450 adult workers’ bees samples (approximately 7-8 bees samples were taken from each station) were collected from 57 shells within the borders of these provinces. The hereditary materials of the samples of the teenage worker bees collected from the clusters are isolated using the technique of phenolclorofrom: isoamilalcol. The DNA samples made by isolations were performed in a series of analyses by taking the images of the agaros gel after the products obtained after PCR were performed in gel electrophoresis. The series analysis data resulted in the material's relativity levels evaluated in the philogenetic tree model.

As a result of this study on bee breeds which have already been spreading in the world and our country, are intended to be determined based upon data obtained from Turkey has been reported that the natural locations of the North Eastern and Eastern Black Sea Caucasian honey’s (A.mellifera caucasica) level of hybridization using the PCR technique and phylogenetic planting method. The genotypic effect of the selection on populations on the species was determined by studying the cytochrome C oxidase I gene of the mitochondrial DNA molecule. The populations of Kars, Ardahan, Erzurum, Artvin and Rize which are located in North Eastern Anatolia and Eastern Black Sea regions are chosen as a goal. As study material approximately 450 adult worker bee samples were collected from 57 bees in the boundaries of these cities (approximately 7-8 bee samples were taken from each station). Hereditary materials of adult bee workers’ samples collected from bees were isolated using fenolklorofrom: isoamylalcohol technique. Isolation of the DNA samples obtained after PCR products, after running on gel electrophoresis, agarose gel images were taken and sequence analyzes were performed. As a result of sequence analysis, the kinship levels of the materials were evaluated in a phylogenetic afforestation model.
