Amaç: Bu çalışmada sezon dışı dönemde, koç spermasının kısa süreli saklanmasında sulandırıcıya farklı oranlarda ilave edilen Galik asitin etkilerinin ortaya konması amaçlandı. Gereç ve Yöntem: Çalışmada Burdur Çallıca kasabasında bulunan özel bir koyun işletmedeki 4 baş Merinos koç (2-3 yaş) kullanıldı. Çalışma süresince koçlardan sperma elektro-ejekülatör yardımıyla haftada iki kez olmak üzere elde edildi. Toplanan ejkülatlardan spermatozoa motilitesi %80>, yoğunluğu 0 mM (kontrol) ile 1.5x10⁹ spermatozoa/ ml üzerinde olanlar pooling yapıldı ve deney grupları oluşturuldu. Deney grupları sırasıyla antioksidan içermeyen (kontrol), 2 mM, 5 mM ve 10 mM dozlarında gallik asit içeren sulandırıcı grupları olacak şekilde belirlendi ve +4 C⁰'de saklandı. Grupların oluşturulmasında sonra 24 saat aralıklar ile 96 saat süresince subjektif motilite (%), morfolojik (%) ve membran bütünlüğü (HOS test, %) bakımından değerlendirildi. Bulgular: Çalışmada 48. saatin sonunda kontrol grubuna (%51.0±2.7) göre en yüksek motilite Gallik asit 2 mM (%72.7±5.3) ve 5 mM (%69.4±4.8) içeren sulandırıcı gruplarından elde edildi (P<0.05). Saklama süresi sonunda (96. saat) gallik asit 2 mM ve gallik asit 5 mM grupları spermatozoa motilitesi kontrol grubuna kıyasla daha fazla koruyucu özellik gösterdi (P<0.05). Buna karşın gallik asit 10 mM membran bütünlüğü bakımından kontrol grubuna göre olumsuz etki gösterdi. (P<0.001). Anormal spermatozoa oranları bakımından her bir zaman dilimi açısından gruplar arasında istatistiki açıdan bir farklılık tespit edilmedi (P>0.05). Ancak, saklama zamanına bağlı olarak anormal spermatozoa oranında artış istatistiksel olarak önemli bulundu (P <0.001). Öneri: Sonuç olarak, sezon dışında koç spermasının kısa süreli saklanması amacıyla sulandırıcıya 2 ve 5 mM dozlarında gallik asitin eklenmesi, motilite oranlarında olumlu etki yapabileceği kanısına varıldı.
Purpose: This study was aimed at revealing the effects of Galic acid, which is added to the moisturizer in different proportions in the non-season period, in the short-term storage of the coach sperm. Tools and Methods: In the study, a private sheep plant located in Burdur Çallıca was used by 4 heads of Merinos coach (2-3 years old). During the study, sperm was obtained from the coaches with the help of an electro-execulator twice a week. The sperm motility of the collected sperm was 80%>, the density of 0 mM (control) with 1.5x109 sperm/ml were pooled and experimental groups were created. The trial groups were determined in such a way that they would be non-antioxidant (control), 2 mM, 5 mM and 10 mM doses of gallic acid and were stored at +4 C0 respectively. After the creation of the groups, 24 hours and 96 hours were evaluated in terms of subjective motility (%), morphological (%) and membrane integrity (HOS test, %). Results: The study at the end of the 48th hour, the highest motility according to the control group (%51.0±2.7) was obtained from the hydrating groups containing 2 mM (%72.7±5.3) and 5 mM (%69.4±4.8) of gallic acid (P<0.05). At the end of the storage period (96. hours) gallic acid 2 mM and gallic acid 5 mM groups showed more protective properties compared to the sperm motility control group (P<0.05). However, gallic acid showed negative effects according to the control group in terms of membrane integrity of 10 mM. (P < 0.001 ) There was no statistical difference between the groups in terms of anormal sperm rate (P>0. and 05). However, an increase in an abnormal sperm rate was statistically significant depending on the storage time (P <0.001). Recommendation: As a result, the addition of gallic acid in doses of 2 and 5 mM to the moisturizer for the purpose of short-term storage of the coach sperm outside the season has been shown to have a positive effect on the motility rates.
