AMAÇ: Bu çalışmada Ankara ilindeki, çeşitli süpermarketlerden temin edilen dana kıyma ve tavuk but örneklerinde S. aureus ve koagülaz negatif stafilokok (KNS)’ların bulunma sıklığı ile bu izolatların biyofilm üretimi ve DNaz aktivitelerinin saptanması amaçlanmıştır. YÖNTEMLER: İncelenen 15 kıyma ve 15 tavuk but örneğinden izole edilen S. aureus ve KNS’lerin cins ve tür düzeyindeki identifikasyonları standart biyokimyasal yöntemler ve API ID 32 Staph (Bio Merieux SA, Marcy-I’Etoile, France) bakteri tanımlama kiti kullanılarak yapılmıştır. Stafilokok türlerinin biyofilm oluşumları Congo Red Agar’da, DNaz aktiviteleri DNaz Agar’ da belirlenmiştir. BULGULAR: İncelenen kıyma örneklerinden 6’sı (% 10,7) S. aureus, 50’ si (% 89,3) KNS olmak üzere toplam 56 stafilokok izolatı elde edilmiştir. Tavuk but örneklerinden toplam 41 KNS izolatı elde edilmiş, S. aureus izole edilememiştir. DNaz aktivitesi kıymadan izole edilen S. aureus izolatlarının tamamında (% 100), KNS’lerin % 58’inde pozitif bulunmuştur. Tavuktan izole edilen KNS’lerin %70,7 ‘si DNaz pozitif bulunmuştur. Biofilm üretimi kıymadan izole edilen S. aureus izolatlarının % 50’sinde, KNS’ lerin % 18’sinde tespit edilmiştir. Tavuktan izole edilen KNS’lerin % 41.5’sinde biyofilm üretimi saptanmıştır. SONUÇ: Stafilokok türlerinin etlere kontaminasyonun bu gıdaların hazırlanması sırasındaki zayıf sanitasyon ve çapraz kontaminasyonun göstergesi olabileceği düşünülmektedir. Bu çalışmada izolatların büyük bir oranda biofilm oluşturdukları ve DNaz aktivitesine sahip odukları gözlenmiştir. DNaz aktivitesi olan ve biofilm oluşturabilen stafilokok türlerinin kontaminasyon ile gıda zincirine girebileceğinin gıda üreticileri tarafından dikkate alınması gerektiği, bu nedenle de hijyenik ve koruyucu önlemlerin alınması gerektiği düşüncesindeyiz.
The aim of this study is to identify the frequency of S. aureus and koagülaz negative staphylococcus (KNS) in the samples of dana cottage and chicken but supplied from various supermarkets in the province of Ankara and the biofilm production of these isolates and DNaz activities. Methods: Identification of the species and species of S. aureus and KNS, isolated from the examined 15 cattle and 15 chicken but examples, was made using standard biochemical methods and API ID 32 Staph (Bio Merieux SA, Marcy-I'Etoile, France) bacterial identification kit. Biophilic formations of staphylococcus species in Congo Red Agar, DNaz activity in DNaz Agar is determined. Results: A total of 56 staphylococcus isolates were obtained, including 6 of the examined samples (10,7%) S. aureus, 50' si (89,3%) KNS. A total of 41 KNS isolates were obtained from the chicken butte samples, S. aureus was not isolated. DNaz activity was found positive in all S. aureus isolates (100%) and 58% of KNSs. 70,7% of KNSs isolated from chicken found DNA positive. Biofilm production is detected in 50% of S. aureus isolates, unprecedented, and in 18% of KNSs. In 41.5 percent of chicken-isolated KNSs, biofilm production was detected. It is believed that the contamination of staphylococcus species in meat may be a indicator of poor sanitation and cross-contamination during the preparation of these foods. In this study, it was observed that isolates made a large proportion of biofilm and had DNA activity. We believe that food producers should consider that staphylococcus species that are active in DNA and can produce biofilm can enter the food chain by contamination, and therefore should take hygienic and protective measures.
