Diyabet Oluşturulan Ratların Böbrek Dokusunda Oksidatif Stres ve Apoptozis Üzerine Perilil Alkolün Koruyucu Etkileri

Bu çalışmada diyabet oluşturulan ratların böbrek dokusunda oksidatif stres ve apoptozis üzerine perilil alkolün etkileri araştırılmıştır. Bu amaçla 40 adet rat eşit olarak rastgele 4 gruba ayrılmıştır. 1.Grup (kontrol), 2. Grup (Diyabet; STZ 50 mg/kg i.p, tek doz), 3. Grup (Perilil Alkol; Perilil Alkol 50 mg/kg/gün p.o, 7 gün), 4. Grup (Diyabet+Perilil Alkol; STZ 50 mg/kg i.p, tek doz+Perilil Alkol 50 mg/kg/gün, p.o 7 gün). Çalışma sonunda böbrek dokusu örneklerinde bazı biyokimyasal parametreler malondialdehit (MDA), glutatyon (GSH), glutatyon peroksidaz (GSH.Px), katalaz (CAT) ve apoptotik indeks markerları olan Bax ve Bcl-2 seviyeleri Western blot analizleri ile ölçülmüştür. Çalışma sonucunda diyabet oluşturulan grupta MDA seviyesi kontrol grubuna göre artmış (P<0.01); GSH seviyesi, GSH-Px ve CAT aktiviteleri ise azalmıştır (P<0.05). Diyabet grubuyla kıyaslandığında, Diyabet+Perilil Alkol grubunda MDA düzeyi azalmış (P<0.01); GSH, GSH-Px ve CAT aktiviteleri ise yükselmiştir (P<0.05). Bax protein düzeyleri Diyabet grubunda kontrol grubuna göre artış göstermiş; fakat Diyabet+Perilil Alkol grubunda azalmıştır (P<0.05). Bcl-2 seviyesi ise Diyabet+Perilil Alkol grubunda artış göstermiştir (P<0.05). Bax/Bcl-2 oranı ise Diyabet grubunda artmış olmasına rağmen (P<0.05), Diyabet+Perilil Alkol grubunda azalmıştır. Sonuç olarak, perilil alkolün diyabetin erken döneminde oluşan oksidatif stresi azalttığı ve antioksidan etkinliği artırdığı, bununla birlikte apoptotik indeks göstergesi olan Bax/Bcl-2 oranını azalttığı tespit edilmiştir.

Abstract:

This study has studied the effects of perylal alcohol on oxidative stress and apoptosis in the kidney tissue of diabetic rats. For this purpose, 40 rates were divided randomly into 4 groups equally. Group 1 (control) and 2. Group (Diabetes; STZ 50 mg/kg i.p, single dose), 3. Group (Perilyl Alcohol; Perilyl Alcohol 50 mg/kg/day p.o, 7 days), 4. Group (Diabetes + Peryl alcohol; STZ 50 mg/kg, single dose + Peryl alcohol 50 mg/kg/day, p.o. 7 days). At the end of the study, some biochemical parameters in kidney tissue samples were measured by Western blot analysis, the Bax and Bcl-2 levels, which are malondialdehitis (MDA), glutation (GSH), glutation peroxidase (GSH.Px), catalase (CAT) and apoptotic index markers. The study resulted in an increase in the MDA level in the group of diabetes created according to the control group (P<0. 01); the level of GSH, while the activity of GSH-Px and CAT has decreased (P<0.05). Compared to the Diabetes group, the MDA levels in the Diabetes+Peryl Alcohol group have decreased (P<0.01) and the GSH, GSH-Px and CAT activities have increased (P<0.05). The Bax protein levels increased according to the control group in the Diabetes group, but it decreased in the Diabetes+Peryl Alcohol group (P<0.05). The level of Bcl-2 has increased in the Diabetes+Peryl Alcohol group (P<0.05). Although the Bax/Bcl-2 rate has increased in the Diabetes group (P<0.05), it has decreased in the Diabetes+Peryl Alcohol group. As a result, it has been found that perylyl alcohol reduces oxidative stress that occurs in the early period of diabetes and increases antioxidant efficacy, while reducing the rate of Bax/Bcl-2, the apoptotic index.
