Amaç: Bu çalışmada, yumurtaya verilen farklı dozlarda BPA?nın iskelet kası gelişimi üzerindeki etkilerinin histolojik metotlarla belirlenmesi amaçlanmaktadır. Gereç ve Yöntem: Isa Brown ırkı sağlıklı ticari anaçlardan elde edilen 310 adet döllü tavuk yumurtası kontrol, taşıyıcı, 50,100 ve 250 ?g/yumurta olmak üzere 5 gruba ayrıldı. Test solüsyonları inkübasyon öncesi yumurta sarısına enjekte edildi. Kuluçkanın 13, 18 ve 21. günlerinde, her gruptan 10 canlı embriyo elde edilinceye kadar rastgele yumurta seçilerek açıldı. Embriyolardan alınan iskelet kası (musculus fibularis longus, musculus sternocoracoideus pectoralis) doku örnekleri %10'luk formol solüsyonunda tespit edildi ve rutin histolojik tekniklerle işlenerek parafinde bloklandı. Kesitler Crossmon?un üçlü boyama yöntemi ile boyandı. PCNA (Proliferating cell nuclear antigen) reaktivitesi ise immünohistokimyasal yöntemle belirlendi. Bulgular: Kas gelişiminin BPA uygulanan gruplarda geri kaldığı ve PCNA pozitif hücre sayısının da daha yüksek olduğu dikkati çekti (p<0.05). Söz konusu bu gruplarda enine bandlaşmanın çok zayıf olduğu ve miyotüp organizasyonunun ise henüz tamamlanmadığı gözlendi. Öneri: BPA'nın iskelet kası dokusunun gelişimi üzerindeki olumsuz etkilerinin ileride hayvanlardan beklenen bazı performans parametrelerinde düşüşe neden olabileceği sonucuna varılmıştır.
Purpose: This study aims to determine the effects of BPA on the development of the bone muscle in different doses given to the egg by histological methods. The Isa Brown breed was divided into 5 groups: 310 fertilized chicken eggs obtained from healthy commercial mothers, controlled, carrier, 50,100 and 250 g/ egg. Test solutions were injected into the pre-incubation egg yellow. On the 13th, 18th and 21st days of the shell, the egg was selected by random selection until 10 living embryos were obtained from each group. Musculus fibularis longus (musculus sternocoracoideus pectoralis) tissue samples taken from embryos were detected in a 10% formol solution and blocked in the paraffin by processing with routine histological techniques. The cuts were painted by the crossmon’s triple coloring method. The PCNA (Proliferating Cell Nuclear Antigen) reactivity was determined by an immune-histochemical method. The findings: It noted that muscle development remained behind in the BPA group and that the number of PCNA positive cells was also higher (p<0.05). It was observed that the bandage in these groups was very weak, and the myotype organization was not yet completed. Recommendation: The negative effects of BPA on the development of the skeletal muscle tissue have been concluded that it could lead to a decline in certain performance parameters expected from animals in the future.
