Amaç: Kronik Miyeloid Lösemi (KML), hematopoetik kök hücreden (HKH) köken alan miyeloproliferatif bir hastalıktır. MikroRNA'lar, transkripsiyon sonrası gen ekspresyonunu düzenlenleyen küçük kodlamayan RNA’lardır. miRNA’lar KML’nin progresyonunda, lösemik kök hücre büyümesi ve tirozin kinaz inhibitörü (TKİ) direncinin gelişmesinde hücre homeostazisini etkilemektedirler. Bu çalışmada KML lösemik hücresi ve KML lösemik kök hücresi (LKH) arasında değişen miRNA ekspresyon profillerinin incelenmesi amaçlanmıştır. Gereç ve yöntem: KML hücre hattı olan K562 hücrelerinden, manyetik hücre ayrımlama (MACS) yöntemi kullanılarak CD34+CD38- lösemik kök hücreleri ayrımlanmıştır. Ayrımlanan LKH’lerin saflığının %85-92 arasında olduğu akım sitometri yöntemi ile gösterilmiştir. K562 ve K562 LKH’leri arasında, gerçek zamanlı kantitatif PCR ile kanser kök hücre ilişkili 84 adet miRNA’nın ekspresyon değişimleri incelenmiştir. Bulgular: K562 ve K562 LKH’leri arasında, kök hücre ilişkili olduğu bilinen 84 adet miRNA’dan 7’sinin anlamlı düzeyde değiştiğini gözledik (P<0,05). K562 LKH’lerinde hsa-miR-29b-3p’nin ekspresyon düzeylerinde artış izlenirken; hsa-miR-320d, hsa-miR-96-5p, hsa-let-7e-5p, hsa-miR-17-5p, hsa-miR-181b-5p hsa-miR-423-5p’da azalma olduğu gözlenmiştir. Sonuç : KML lösemik hücreleri ile KML LKH’leri arasında proliferasyon, eritroid farklılaşma, kendi kendini yenileme ve apoptoz sürecinde rol alan miRNA’lar ve hedef genlerinin ekspresyonlarındaki değişim, hastalığın ilerlemesinde miRNA’larında rol oynayabileceğini göstermektedir. Bu nedenle KML LKH’lerine özgü miRNA’ların hastalığın progresyonunun ve TKI direncinin önlenmesi için yeni terapötik stratejilerin geliştirilmesinde hedef moleküller olabileceğini düşünmekteyiz.
Chronic Myeloid Lusemia (KML) is a myeloproliferative disease that originates from the hematopoetic root cell (HKH). MicroRNAs are small non-coded RNAs that regulate the post-transcription gen expression. MiRNAs affect cell homeostasis in the progression of KML, the growth of the leukemic root cell and the development of the resistance to the thyrocin kinase inhibitor (TI). This study aims to study the miRNA expression profiles that vary between the KML leukemic cell and the KML leukemic root cell (LKH). Tools and methods: From the K562 cells, the KML cell line, the CD34+CD38-lecemic root cells are separated using the magnetic cell separation (MACS) method. It is demonstrated by the flow cytometry method where the purity of separated LKHs is between 85-92%. Between K562 and K562 LKHs, expression changes of 84 miRNAs associated with cancer root cells in real-time quantitative PCRs have been studied. Results: Between K562 and K562 LKHs, we observed that 7 of the 84 miRNAs known to be associated with root cells changed significantly (P<0,05). In the K562 LKH there is an increase in expressions of hsa-miR-29b-3p; there is a decrease in hsa-miR-320d, hsa-miR-96-5p, hsa-let-7e-5p, hsa-miR-17-5p, hsa-miR-181b-5p, hsa-miR-423-5p. The result: The proliferation between the KML leukemic cells and the KML LKHs, erythroid differentiation, the change in the expressions of myRNAs and target genes that play a role in the process of self-renovation and apoptosis, indicates that the progression of the disease can play a role in myRNAs. Therefore, we consider that myRNAs characterized by KML LKH may be targeted molecules in the development of new therapeutic strategies to prevent the progression of the disease and resistance to TKI.
