ÖZET Amaç: Bu çalışma, sıçanlarda deneysel olarak oluşturulan böbrek iskemi-reperfüzyon hasarında güçlü antioksidan olan Klorojenik asitin antiinflamatuar ve antioksidan etkilerini araştırmayı hedeflemektedir. Gereç ve Yöntemler: Otuz altı adet erişkin, Wistar albino cinsi dişi sıçan, rastgele dört gruba ayrıldı. Sham grubundaki sıçanlar (Grup I), sırt bölgesinden açılarak sağ nefrektomi yapıldı. İskemi-reperfüzyon grubunda (Grup II), sıçanların sırt bölgesi açılıp sağ nefrektomi yapıldıktan sonra sol renal arter klemplendi. Grup III ‘te 5 mg/kg Klorojenik asit iskemiden 10 dk önce ve reperfüzyondan 10 dk önce olmak üzere iki doz şeklinde intraperitoneal olarak uygulandı. Sol böbreğe 1 saatlik iskemiden sonra 24 saatlik reperfüzyon uygulandı. Grup IV’te Grup III’ten farklı olarak 10 mg/kg Klorojenik asit uygulandı. Böbrek dokusunda total antioksidan düzeyi (TAS), total oksidan düzeyi (TOS), süperoksit dismutaz (SOD), malondialdehit (MDA) ve myeloperoksidaz (MPO) seviyeleri spektrofotometrik yöntemlerle ölçüldü. Oksidatif stres indeksi (OSI) hesaplandı. Bulgular: Grup II’de Grup I’e kıyasla TOS, MDA ve MPO düzeylerinin arttığı; TAS ve SOD düzeylerinin ise azaldığı görüldü (her bir parametre için p<0.001). Klorojenik asit uygulanan gruplarda böbrek TOS (p<0.001), MDA (p<0.001) ve MPO (p<0.001) düzeylerinde azalma; TAS (p<0.05) ve SOD (p<0.001) değerlerinde ise artış tespit edildi. Sonuç: Böbrek İ/R hasarında oksidatif stresin önemli rol oynadığı ve bu hasara karşı Klorojenik asit uygulamasının oksidatif hasarı belirgin olarak azalttığı görüldü.
This study aims to investigate the anti-inflammatory and antioxidant effects of chlorogenic acid, which is a powerful antioxidant in kidney ischemic-reperfusion damage experimentally created in rats. Tools and Methods: Thirty-six adult, Wistar albino sex female rats, randomly divided into four groups. The rats in the Sham group (Group I) were opened from the back area to the right nefrectomy. In the Iskemi-reperfusion group (Group II), the back area of the rats was opened and the left renal artery clamped after the right nefrectomy was performed. In Group III, 5 mg/kg of chlorogenic acid was administered intraperitoneally in the form of two doses, 10 minutes before and 10 minutes before reperfusion. A 24-hour reperfusion was applied to the left kidney after 1 hour ischemic. In Group IV, 10 mg/kg of chlorogenic acid was administered, unlike Group III. The total antioxidant levels (TAS), total oxidant levels (TOS), superoxid dismutaz (SOD), malondialdehitis (MDA) and myeloperoxidase (MPO) levels in the kidney tissue were measured by spectrophotometric methods. The oxidative stress index (OSI) is calculated. Results: In Group II, TOS, MDA and MPO levels increased compared to Group I, while TAS and SOD levels decreased (p<0.001 for each parameter). A decrease in the levels of kidney TOS (p<0.001), MDA (p<0.001) and MPO (p<0.001) in the groups applied chlorogenic acid, and an increase in the TAS (p<0.05) and SOD (p<0.001) values. The result is that oxidative stress plays an important role in kidney I/R damage and the application of chlorogenic acid against this damage has shown to significantly reduce the oxidative damage.
