Purpose: Determination of micobacteria at species level, the distinction of pathogenic species from non-pathogenic species, the choice of the appropriate treatment regime and the collection of epidemic data are important. Traditional methods and laboratory diagnosis require faster, sensitive and reliable methods to identify micobacteria that are time-consuming and difficult. The aim of this study is to determine the suitability of the hsp65 Polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) method for the routine laboratory use. Method: A total of 141 micobacterial insulates were produced in sequence from 1632 patients sent to the Medical Microbiology Laboratory in a three-year period. In the identification of isolates, the MGIT PNB, niasin test and molecular methods of the hsp65 PCR-RFLP method were used. Conventional methods of micobacterial reproduction in the culture identify 138 M. tuberculosis complex (MTBC) and three non-tuberculosis micobacteria (TDM). Hsp65 PCR-RFLP method is defined as 137 isolat MTBC, four isolat TDM. The conventional method is PNB-sensitive because it is determined as TDM by a isolated hsp65 method that is considered MTBC. In the identification of non-tuberculosis micobacteria with the hsp65 PCR-RFLP method, an isolate M. abcessus is defined as three isolates M. avium complex. The result: In our study, the hsp65 PCR-RFLP method, which enables the identification of micobacteria including TDMs, has come to the conclusion that it is a cheap, easy and routine method to provide clinical information quickly.
