Bu çalışmada asetik asit ile oluşturulmuş deneysel ülseratif kolitis modelinde zingeron'un etkileri araştırıldı. Bu amaçla her grupta 7 sıçan olacak şekilde 4 grupta çalışma yapıldı. Gruplar; kontrol (serum fizyolojik - intrarektal), kolitis (%3 asetik asit - intrarektal), sülfasalazin (50 mg/kg) + kolitis (%3 asetik asit - intrarektal) ve zingeron (20 mg/kg) + kolitis (%3 asetik asit - intrarektal) olacak şekilde oluşturuldu. Elde edilen verilere göre, kolitis oluşturulan grupta kontrol grubuna göre malondialdehit (MDA) seviyeleri artış göstermiş (P<0.01), redukte glutatyon (GSH) seviyeleri (P<0.05), glutatyon peroksidaz (GSH.Px) (P<0.05) ve katalaz (CAT) (P<0.001) aktiviteleri ise azalma göstermiştir. Bununla birlikte zingeron + kolitis grubunda ise kolitis grubuna göre, MDA seviyeleri azalmış (P<0.01), GSH seviyeleri (P<0.05) ve GSH.Px aktiviteleri (P<0.05) ise artış göstermiştir. Sülfasalazin ile kolitis uygulanan grupta ise sadece kolitis oluşturulan gruba göre MDA seviyeleri azalmış (P<0.01), GSH seviyeleri (P<0.05) ve GSH.Px (P<0.05) ve CAT (P<0.01) aktiviteleri ise artış göstermiştir. Histopatolojik analizlerde kolitis oluşturulan sıçanların kolonlarında yoğun epitel ve kript kaybı ile birlikte mukozadan serozaya uzanan nekroz ve diffuz yangısal hücre infiltrasyonu görüldü. Koruyucu amaçla kolitis ile birlikte zingeron ve sülfasalazin verilen gruplarda kolitis grubuna oranla daha hafif derecede mukoza ve submukozda sınırlı nekroz ve yangısal hücre infiltrasyonu dikkat çekti. Kontrol grubu dışındaki gruplarda farklı derecelerde eozinofil lökosit infiltrasyonu (eozinofilik kolitis) gözlendi. Sonuç olarak zingeron'un asetik asit ile oluşturulan deneysel ülseratif kolitis modelinde koruyucu etkilerinin olabileceği görülmüştür.
In this study, the effects of zingeron in the experimental ulcerative colitis model created with acetic acid were studied. For this purpose, there were four groups of rats in each group. The groups were formed in such a way that control (serum physiological - intrarectal), colitis (3% acetic acid - intrarectal), sulfasalazine (50 mg/kg) + colitis (3% acetic acid - intrarectal) and zingeron (20 mg/kg) + colitis (3% acetic acid - intrarectal) will be. According to the data obtained, the levels of malondialdehitis (MDA) increased (P<0.01), the levels of reduced glutathion (GSH) (P<0.05), glutathion peroxidase (GSH.Px) (P<0.05) and catalase (CAT) (P<0.001) in the group created. However, in the Zingeron + colitis group, the MDA levels have decreased (P<0.01), the GSH levels (P<0.05) and the GSH.Px activity (P<0.05) have increased. In the group applied with sulfasalazine and colitis, MDA levels (P<0.01), GSH levels (P<0.05) and GSH.Px (P<0.05) and CAT (P<0.01) increased compared to the group created with colitis. In histopathological analyses, necrosis and diffuse inflammatory cell infiltration, which extends from mucous to serosis, along with intense epithelial and cryptal loss in the colons of colitis-created rats, were observed. For protective purposes, cingeron and sulfasalazine were given in groups with colitis, with a mild infiltration of necrosis and fire cells limited in the mucosa and submucosa compared to the colitis group. In groups outside the control group, eozinofil lacosite infiltration (eozinofil colitis) was observed at different degrees. As a result, it has been shown that zingeron may have protective effects in the experimental ulcerative colitis model created with acetic acid.
