Amaç: Kistik ekinokokkozis (KE), Echinococcus granulosus sensu lato’nun (s.I.) larval formu tarafından oluşturulan; insanları, çiftlik hayvanları ve yaban hayvanlarını etkileyen zoonoz karakterli bir enfeksiyondur. Son yıllarda yapılan moleküler ve taksonomik çalışmalar neticesinde, KE’ye beş kriptik türün bir kompleksi olan Echinococcus granulosus s.l.’nun yol açtığı kabul edilmiştir. Bu çalışmada, hidatid kist ile doğal enfekte bir yaban domuzu ve bir katırdan elde edilen kist materyallerinin morfolojik ve moleküler karakterizasyonları yapılmıştır. Yöntemler: gDNA izolasyonundan sonra spesifik primerler kullanılarak, mitokondrial sitokrom oksidaz subunit 1 (mt-CO1) gen bölgesi polimeraz zincir reaksiyon (PZR) ile çoğaltılmıştır. Amplifiye olan mt-CO1 PZR ürünleri saflaştırılıp, tek yönlü DNA dizi analizi gerçekleştirilmiştir. Bulgular: GenBank’taki referans sekanslar ile kıyaslanması sonucunda, hidatid kist izolatlarının mt-CO1 gen bölgesi kısmi sekanslarının E. granulosus sensu stricto (G1-G3) sekanslarıyla %100 benzerlik gösterdiği belirlenmiştir. Sonuç: Bu çalışma, Türkiye’de yaban domuzlarında Echinococcus türlerinin moleküler karakterizasyonunun belirlenmesine yönelik yapılmış ilk çalışmadır.

In this study, morphological and molecular characteristics of a wild pig infected naturally with hydatide cysts and cyst materials obtained from a cataracts were made. Methods: using specific primates after gDNA isolation, the mitochondrial cytochrome oxidase subunit 1 (mt-CO1) is multiplied with the gene area polymerase chain reaction (PZR). The extended mt-CO1 PZR products were purified and one-way DNA series analysis was carried out. Results: As a result of a comparison with the reference sequences in GenBank, it was found that the part sequences of the hydatid cyst isolat in the mt-CO1 gen area were 100% similar to the sequences of E. granulosus sensu stricto (G1-G3). This study is the first study in Turkey aimed at determining the molecular characterization of Echinococcus species in wild pigs.
