Gıdalarda Lateral Akış Testi ile Listeria Analizi

Listeria monocytogenesgıda kökenli önemli bir patojendir ve gıda, çevre ve klinik örneklerde yoğun bir şekilde analizi yapılmaktadır. Bu çalışmada 50 gıdada selektif zenginleştirme sonrası Singlepath® Listeria hızlı test kiti kullanılarak Listeria spp. analizi yapılmıştır. Ayrıca sırası ile 1/2 ve 1/1 Fraser Broth kullanılarak yapılan ön zenginleştirme ve selektif zenginleştirme aşamalarından sonra PALCAM Agar’a sürme yapılmış, izole edilen tipik koloniler tanımlanmıştır. Çalışma sonuçlarına göre, örneklerin %42’sinden Listeria spp. izole edilmiştir. L. monocytogenes %20 (10/50) oranla en yaygın olan tür iken, bunu %16 (8/50) ile L. innocua izlemiştir. Fraser Broth’ta eskulin parçalanmasına bağlı olarak renk değişimi ile karara varılmasının yanıltıcı olduğu görülmüştür. Zenginleştirmenin farklı aşamalarından PALCAM Agar’a yapılan ekimler sonunda farklı Listeria türleri elde edilebildiği gibi sadece ön zenginleştirme ya da sadece selektif zenginleştirme aşamasında L. monocytogenes de izole edilebilmiştir. Bulgular, standart analiz yöntemlerine tümüyle uyulması gerektiğini göstermiştir.

Listeria monocytogenes is an important pathogen of food origin and is thoroughly analyzed in food, environment and clinical samples.In this study, after selective enrichment in 50 foods, Singlepath Listeria uses a quick test kit with Listeria spp.®The analysis has been done.Also, typical isolated colonies are identified, which have been driven to PALCAM Agar after the pre-enrichment and selective enrichment stages made using 1/2 and 1/1 Fraser Broth in order.According to the results of the study, 42% of the samples were Listeria spp.It is isolated.L. monocytogenes was the most common species with a rate of 20% (10/50), while it was followed by L. innocua with a rate of 16% (8/50).In Fraser Broth, it has been seen as misleading to make a decision with a change of color due to the breakdown of the cluster. From the different stages of enrichment to PALCAM Agar, different Listeria species can be achieved at the end of the decade, and L. monocytogenes can be isolated only in the pre-enrichment or only in the selectively enrichment phase. The findings showed that the standard analysis methods should be fully followed.
