Tokat Kabakgil Üretim Alanlarında Enfeksiyon Oluşturan Virüslerin Belirlenmesi

Karadeniz ve İç Anadolu bölgeleri arasında geçit kuşağında yer alan Tokat Türkiye sebze üretiminde önemli bir paya sahip olup, yörede üreticiler için sebzeler önemli bir gelir kaynağını oluşturmaktadır. Ancak son yıllarda yörede artan virüs enfeksiyonlarından dolayı sebze üretiminde problemler yaşanmaktadır. Bu çalışmada Tokat ilinde yoğun olarak kabakgil (karpuz, kavun, kabak ve hıyar) yetiştirilen alanlarda sorun olan virüslerin belirlenmesi amaçlanmıştır. Bu amaçla Tokat ilinin Merkez, Erbaa, Niksar, Pazar ve Turhal ilçelerindeki kabakgil alanlarında 2014 yılında gerçekleştirilen surveylerde; yapraklarda iplikleşme, kabarcıklaşma ve mozaik; bitkinin genelinde rozetleşme, gelişme geriliği ve bodurlaşma; meyvede şekil bozukluğu belirtisi gösteren bitkilerden toplam 571 yaprak örneği toplanmıştır. Alınan örnekler, Kabak sarı mozaik virüsü (Zucchini yellow mosaic virus; ZYMV), Hıyar mozaik virüsü (Cucumber mosaic virus; CMV), Karpuz mozaik virüsü (Watermelon mosaic virus; WMV), Papaya halkalı leke virüsü (Papaya ring spot virus; PRSV), Kabak mozaik virüsü (Squash mosaic virus; SqMV), Domates mozaik virüsü (Tomato mosaic virus; ToMV), Patates Y virüsü (Potato virus Y; PVY) ve Tütün mozayik virüsü (Tobacco mosaic virus; TMV)’ne spesifik antiserumlar kullanılarak double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) yöntemi ile test edilmiştir. Test sonuçlarına göre 571 örneğin  %37’sinin WMV, %12’sinin ZYMV, %11’inin CMV, %3,4’ünün PRSV ve %0,2’sinin SqMV ile enfekteli olduğu saptanmıştır. ToMV, TMV ve PVY için toplam 287 kabakgil örneği test edilmiş ve yaklaşık %14’ünün ToMV ile bulaşık olduğu belirlenmiştir. TMV ve PVY etmenlerine rastlanmamıştır. Tüm kabakgil türleri arasında virüsle bulaşıklık  oranları; kabak  %74, kavunda %72,4,  karpuz  %47,7  ve hıyar %31,7 olarak belirlenmiştir.

Located in the passage circle between the Black Sea and the Inner Anatolian regions, Tokat Turkey has an important role in vegetable production, and vegetables are an important source of income for producers in the area. However, in recent years there have been problems in vegetable production due to increased viral infections in the area. In this study, the aim is to identify viruses that have problems in areas that are widely grown in the Tokat province (carpuz, cabbage, cabbage and cabbage). For this purpose, in the survey conducted in 2014 in the cabbage areas in the Central, Erbaa, Niksar, Pazar and Turhal districts of Tokat, a total of 571 sheet samples of plants that show a sign of shape disorder in the fruit have been collected in the leaves; sliding, bubbling and mosaic; rosetization, development tension and bodurization throughout the plant. Obtained examples include Cucchini yellow mosaic virus (ZYMV), Cucumber mosaic virus (Cucumber mosaic virus; CMV), Cucumber mosaic virus (Watermelon mosaic virus; WMV), Papaya curved spike virus (Papaya ring spot virus; PRSV), Cucumber mosaic virus (Squash mosaic virus; SqMV), Domates mosaic virus (Tomato mosaic virus; ToMV), Patates Y virus (Potato virus Y; PVY) and Tobacco mosaic virus (Tobacco mosaic virus; TMV) specific use of the antibodies is directed with the double-linked sandwich-en-linked immunosystem (ASB) test. According to the results of the test, 571 of the 37% were infected with WMV, 12% were infected with ZYMV, 11% were infected with CMV, 3.4% were infected with PRSV and 0.2% were infected with SqMV. A total of 287 shell samples for ToMV, TMV and PVY have been tested and 14% have been found to be washed with ToMV. TMV and PVY are not found. The rates of viral infection among all species of pimples are determined: pimples are 74, pimples are 72.4, pimples are 47.7 and pimples are 31.7%.
