Abstract:

Purpose: The biophilic ability that Staphylococcus aureus regulates through the icaA and icaR genes makes it difficult to fight the infections they create. In this study, the aim was to study in-vitro the effects of Lactobacillus acidophilus and Lactobacillus casei, methysillin-sensitive (MSSA) and methysillin-resistant (MRSA) S. aureus juices on icaA and icaR gen expressions. Tools and Methods: 6 tubes are prepared for the control of IcaA and icaR gen expressions. It was incubated at 37°C in an atmosphere with 5% CO2. In the 6th, 12th, and 24th hours of the incubation, RNA isolations were made. After cDNA synthesis, ICA, icaR and 16SrRNA genes were studied with real-time PCR with specific primes. The results were analyzed according to the Delta Delta Ct method. Results: S. aureus ATCC 43300 (MRSA) and S. aureus ATCC 29213 (MSSA) icaA genes and icaR expressions, after encounter with L. acidophilus and L. casei, showed down regulation and upregulation accordingly. It was observed that the levels of icaA and icaR genes identified at 12h and 24h were similar in both MRSA and MSSA doses. Recommendation: As a result, our study is the first study that studies the biofilm effects of probiotic-effective L. acidophilus and L. casei juices of S. aureus juices and icaA and icaR gen expressions with real-time PCR method. L. acidophilus and L. casei have been found to be able to change the icaA and icaR genes expression by influencing the MRSA and MSSA mixture in the in-vitro environment. The probiotic effect of these juices is the antibiotic properties of S. It is believed that there may be a new option to fight aureus infections.
