Bu çalışmanın amacı, kontaminasyon öncesi aside adaptasyon kazandırılmış veya kazandırılmamış olan E. coli O157:H7 ve Listeria monocytogenes'in Şavak tulum peynirinde olgunlaşma süresince boyunca yaşam kabiliyetlerini incelemektir. Olgunlaştırma aşamasına alınacak olan Şavak tulum peynirleri aside adapte edilmiş ve edilmemiş E. coli O157:H7 ve Listeria monocytogenes ile kontamine edildi ve 120 günlük olgunlaştırma periyodu boyunca sayımları yapıldı. Her iki patojen bakteri sayısı için de tüm olgunlaştırma periyodu boyunca aside adapte edilmiş ve aside adapte edilmemiş gruplar arasındaki farkın önemsiz olduğu tespit edildi (P>0.05). Olgunlaşmanın ilk günü kontamine edilen peynirlerde aside adapte ve adapte edilmemiş E. coli O157:H7 sayıları sırayla, 5.39 ve 5.70 log10 kob/g olarak tespit edilmişken, olgunlaşmanın 120. günü sırasıyla 3.64 ve 4.17 log10 kob/g olarak saptandı. Olgunlaşmanın 0. gününde aside adapte ve adapte edilmemiş L. monocytogenes sayısı sırasıyla 5.95 ve 6.84 log10 kob/g olarak belirlenmişken, olgunlaşmanın son günü sırasıyla 3.53 ve 3.01 log10 kob/g olarak saptanmıştır. Sonuç olarak; olgunlaşmanın başlangıcında deneysel olarak kontamine edilmiş Şavak tulum peyniri örneklerinde 120 günlük olgunlaşma süresi boyunca L. monocytogenes ve E. coli O157:H7'nin yaşama kabiliyeti üzerine asit adaptasyonun etkisinin önemli olmadığı tespit edilmiştir. Olgunlaşma periyodu sonunda bu patojen mikroorganizmaların sayılarının ortalama 2.0 log azalmasına rağmen sayılarının enfektif dozlarının çok üstünde kaldığı saptanmıştır. Bu durum, Şavak tulum peyniri gibi çiğ sütten yapılan ve uzun süre düşük sıcaklıklarda olgunlaştırılan peynirlerin L. monocytogenes ve E. coli ile kontamine olması durumunda halk sağlığı yönünden ciddi riskler meydana getirebileceğini göstermektedir.
The aim of this study is to study the lifecapacities of E. coli O157:H7 and Listeria monocytogenes, which have been or have not been given pre-contamination acid adaptation during maturation. The Cheesecake cheesecake, which is to be taken into the maturity stage, was acid-adapted and not contaminated with E. coli O157:H7 and Listeria monocytogenes, and counted throughout the 120-day maturity period. For the number of both pathogens, the difference between the acid-adapted and non-acid-adapted groups during the entire maturity period was also unimportant (P>0.05). On the first day of maturity, the acid-adapted and unadapted E. coli O157:H7 numbers are in order, 5.39 and 5. 70 log10 was identified as kob/g, while the 120th day of maturity was identified as 3.64 and 4.17 log10 kob/g respectively. The number of acid-adapted and unadapted L. monocytogenes was determined at 5.95 and 6.84 log10 kob/g, while the last day of maturation was determined at 3.53 and 3.01 log10 kob/g, respectively. As a result, the acid adaptation effect on the lifecapacity of L. monocytogenes and E. coli O157:H7 during the 120-day maturation period was not significant in the samples of sheep cheese experimentally contaminated at the beginning of maturation. At the end of the maturity period, the number of these pathogenic microorganisms has been reduced by an average of 2.0 logs, although the number of these infectious microorganisms has remained far above the infectious doses. This situation shows that cheese made from raw milk and mature at low temperatures for a long time can cause serious health risks if contaminated with L. monocytogenes and E. coli.
