Bu çalışmanın amacı, tavşan spermasının kısa süreli saklanması süresince (0., 6., 12., ve 24. saat) taxifolinin spermatolojik parametreler üzerine etkisini araştırmaktır. Araştırmada sperma vericisi olarak 8 adet ergin Yeni Zelanda tavşanı kullanıldı. Sperma suni vajen yardımıyla haftada üç kez alındı. Tavşanlardan alınan ejakulatlar 37oC’de değerlendirildikten sonra karıştırıldı. Sperma örnekleri 4 ayrı gruba ayrıldı. Daha sonra sperma numuneleri Tris (K-Tris), Tris+10 µl taxifolin (10 TT), Tris+100 µl taxifolin (100 TT) ve Tris+500 µl taxifolin (500 TT) ile sulandırıldı. Sulandırılmış sperma numuneleri 4oC’ de 24 saat süreyle kısa süreli saklandı. Saklama süresinin ardından bazı spermatolojik parametreler (motilite, akrozom anomali oranı, toplam anormal spermatozoa oranı ve HOST) yönünden analiz edildi. Deneysel çalışmalar 10 kez tekrar edildi. Çalışmanın sonucunda, tüm deneysel gruplarda 24. saat sonunda anormal spermatozoa oranında artış ve plazma membran bütünlüğü ile motilite oranında düşme saptandı. Kontrol grubu verileriyle karşılaştırıldığında 100 TT grubu 6. (%82.50), 12. (%80) ve 24. (%77.50) saatlerde istatistiki olarak en yüksek motilite oranını verdi (P<0.001). Kısa süreli saklamanın 24. saatinde 100 µl taxifolin ilavesinin diğer gruplara kıyasla akrozomal morfolojik yapının bütünlüğünü istatistiki açıdan önemli derecede koruduğu ve akrozom anomalisi oranı (%7) ile toplam spermatozoa anomali oranını (%8.50) düşürdüğü ve plazma membran bütünlüğünü de istatistiki olarak önemli derecede koruduğu saptandı (P<0.001). Sonuç olarak, 100 µl taxifolin ilavesinin tavşan spermasının kısa süreli saklanmasında etkin koruyucu bir rol oynadığı tespit edildi.
The aim of this study is to investigate the effect of taxifoline on the sperm parameters during the short-term storage period (0., 6, 12, and 24 hours) of rabbit sperm. In the study, eight young New Zealand rabbits were used as sperm donors. The sperm was taken three times a week with the help of the artificial vagina. The ejaculates taken from the rabbits were mixed after being assessed at 37oC. Sperm samples are divided into 4 separate groups. The sperm samples were then diluted with Tris (K-Tris), Tris+10 μl taxifolin (10 TT), Tris+100 μl taxifolin (100 TT) and Tris+500 μl taxifolin (500 TT). The moisturized sperm samples were stored at 4oC for a short period of 24 hours. After the storage period, some spermatological parameters were analyzed in the direction (motility, acrosom anomaly rate, total abnormal sperm rate and HOST). Experimental studies have been repeated 10 times. The study found an increase in the rate of abnormal sperm at the end of 24 hours in all experimental groups and a decrease in the rate of motility with the integrity of the plasma membrane. In comparison with the data of the control group, 100 TT group is 6. (% 82.50 ) and 12. 80 and 24%. (77.50%) gave the highest statistically motility rate in hours (P<0.001). In 24 hours of short-term storage, the 100 μl taxifolin supplement, compared to other groups, statistically significantly preserves the integrity of the acrosomal morphological structure and acrosomal anomaly rate (%7) with the total sperm anomaly rate (%8). It was found that it decreased and the integrity of the plasma membrane was also statistically significantly
The aim of this study was to investigate the effect of taxifolin on spermatological parameters of rabbit semen during liquid storage (0, 6, 12, and 24th h). In this study, eight adult New Zealand male rabbits were used as semen donors. Semen was taken with the aid of artificial vagina three times a week. Ejaculates collected from bucks were evaluated and pooled at 37oC. Semen samples were divided into 4 groups then were diluted with Tris (K-Tris), Tris + 10 µl taxifolin (10 TT), Tris + 100 µl taxifolin (100 TT) and Tris + 500 µl taxifolin (500 TT). The diluted semen samples were liquid stored at 4°C up to 24th h. After the storage period, some spermatological parameters (the percentage of motility, acrosome - total spermatozoa abnormality and HOST) were analyzed. Experimental studies were replicated 10 times. As a result of the study, the percentage of abnormal spermatozoa increased and plasma membrane integrity and motility rate decreased at the end of 24th h in all experimental groups. When compared to the control group data, 100 µl taxifolin addition gave the highest motility rate at 6th (82.50%), 12th (80%) and 24th (77.50%) hour (P <0.001). At the 24th hour of liquid storage, 100 µl taxifolin addition was found to protect statistically significantly the integrity of acrosomal structure and plasma membrane compared to other groups. In addition, it was determined that the percentage of acrosome (7%) and total spermatozoa abnormality (8.50%) was low in the 100 TT group when compared to other groups (P <0.001). In conclusion, it was found that addition of 100 µl taxifolin plays effective protective role on short-term storage of rabbit semen.
