Entamoeba histolytica is found throughout the world but prevalence changes from country to country, and region to region. Amoebiosis is relatively common in our country and is an ongoing community health problem. In the present study, we aimed to determine the comparative diagnosis of amoebiosis with nativelugol, sedimentation and trichrome stain methods in patients applying to Erciyes University, Medical Faculty, Parasitology Department. In this study, 1000 stool samples were analyzed. Of the total 1000, 11 (1,1%) stool samples were examined directly with saline-iodine preparations and 13 (1,3%) examined after sedimentations were found positive for E. histolytica/E. dispar. Nine (0,9%) stool samples were examined directly with trichrome staining method and 12 (1,2%) were examined in the same method after sedimentation, and were found positive for E. histolytica/E. dispar. Nativ-lugol is the easiest, least timeconsuming and the cheapest method for the diagnosis of amoebiosis. The formol-ethil asetat concentration method increases the occurrence of E. histolytica/E. dispar cysts, whereas the trichrome staining method precisely differentiates E. histolytica/E. dispar from other organisms. As a result, trichrome staining method is more beneficial in specific diagnosis
