Sigara Dumanına Maruz Kalan Sıçanlarda Lipit Profili ve Karaciğer Fonksiyon Testleri Üzerine L -NAME ve Vitamin E' nin Etkileri

Sigara milyonlarca insan tarafından kullanılmakta olup çevreye ve diğer canlılara doğrudan zarar verir. İçindeki binlerce zararlı madde nedeniyle farklı metabolizmalar üzerinden karaciğer, kalp ve akciğer gibi birçok organı tahrip eder. Bu çalışmada ticari sigaradan elde edilen sigara dumanına inhalasyon yoluyla maruz kalmadan kaynaklanan oksidatif stres sonucunda değişen bazı kan parametresi düzeylerinin Vitamin E ve L-NAME uygulaması ile değerlendirmesi amaçlanmıştır. Çalışmada, 45 erkek Wistar albino sıçan, her grupta 9 sıçan olmak üzere beş gruba ayrıldı. Sigara dumanı özel kafeslerde inhalasyon ile sağlanırken, 200 mg/kg vücut ağırlığı (VA) Vitamin E ve 50 mg/kg VA L-NAME intraperitoneal olarak 42 gün boyunca uygulandı. 43. günde, sıçanlardan kan örnekleri alındı. Kan örneklerinde uygun işlemlerden sonra alanin aminotransferaz (ALT), aspartat aminotransferaz (AST), trigliserit, yüksek yoğunluklu lipoprotein kolesterol (HDL-C), düşük yoğunluklu lipoprotein kolesterol (LDL-C) ve kolesterol düzeyleri incelendi. Sigara dumanı grubunda ALT, AST, trigliserid, kolesterol ve LDL-C düzeylerinde kontrol grubuna göre anlamlı düzeyde artış gözlenirken (p<0.05), HDL-C düzeyinde ise anlamlı derecede düşüş gözlendi (p<0.05). Tedavide grupları değerlendirildiğinde ALT, AST, trigliserit, total kolesterol ve LDL-C düzeylerinin azaldığı ve HDL-C düzeylerinin ise arttığı sonucuna varılmıştır (p<0,05). Bu çalışmanın sonucunda, sigara dumanına maruz kalmanın bazı kan parametrelerinde olumsuz değişikliklere neden olduğu bulunmuştur. Bu olumsuz değişikliklerin ise L-NAME ve Vitamin E uygulaması ile normalleştirilebileceği gözlendi.

Abstract:

Smoking is used by millions of people and is directly harmful to the environment and other animals. It destroys many organs, such as the liver, heart and liver, through different metabolisms due to thousands of harmful substances in it. This study aims to evaluate certain levels of blood parameters that change as a result of oxidative stress resulting from the exposure to the cigarette smoke derived from commercial cigarette through inhalation with the application of Vitamin E and L-NAME. In the study, 45 male Wistar albino rats were divided into five groups, including 9 rats in each group. While smoking smoke is supplied by inhalation in special cafes, 200 mg/kg body weight (VA) Vitamin E and 50 mg/kg VA L-NAME were intraperitoneally administered for 42 days. On the 43th day, blood samples were taken from mice. After appropriate processes in blood samples, the levels of alanin aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride, high-intensity lipoprotein cholesterol (HDL-C), low-intensity lipoprotein cholesterol (LDL-C) and cholesterol were studied. In the smoke group, the levels of ALT, AST, triglycerides, cholesterol and LDL-C were observed significantly increased (p<0.05), while the levels of HDL-C were observed significantly decreased (p<0.05). When the treatment groups were evaluated, the result was that ALT, AST, triglyceride, total cholesterol and LDL-C levels fell and HDL-C levels increased (p<0,05). This study found that exposure to cigarette smoke caused negative changes in some blood parameters. These negative changes could be normalized by the application of L-NAME and Vitamin E.

In this study, it was aimed to evaluate the levels of certain blood parameters that changed as a result of oxidative stress resulting from exposure to cigarette smoke from commercial cigaratte through inhalation with the application of Vitamin E and L-NAME.  In the study, 45 male Wistar albino rats were divided into five group and each group had 9 rats. Cigaratte smoke was administered by inhalation in special cages, 200 mg/kg-BW Vitamin E and 50 mg/kg-BW L-NAME was administered by intraperitoneal during 42 days. On day 43, blood samples were taken from the rats. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), Triglyceride, high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C) and cholesterol levels were examined in blood samples. In the cigaratte smoke group; It was observed significant increase in LDL-C, triglyceride, ALT, AST and total cholesterol levels(p <0.05) and significant decrease HDL-C levels compared to the control group(p <0.05). ALT, AST, Triglyceride, LDL-C and cholesterol levels decreased and HDL-C level increased in treatment groups(p<0,05). Exposure to cigaratte smoke has been found to cause adverse changes in some blood parameters. It was observed that these adverse changes could be normalized by L-NAME and Vitamin E application.
