Türkiye, dünyanın üç önemli gen merkezinin kesisme noktası olan bir bölgede bulunması nedeniyle bitki genetik kaynakları açısından büyük bir potansiyele sahiptir. Ülkemizin tür zenginliği içinde doğal çiçek soğanlarının ayrı bir yeri vardır. Bu bitkiler yüzyıllardır tıbbi amaçlarla kullanılmasına karşın, kış aylarında çiçeklenmeleri nedeniyle genis ölçüde bahçelerde süs bitkisi olarak da kullanılmaktadır. Yaprak lekesi, kök çürüklüğü ve solgunluk etmenleri gibi birçok patojeni bünyesinde barındıran soğanlı süs bitkilerinde yaygın olarak görülen Fusarium solgunlukları her yıl büyük ekonomik kayıplara neden olmaktadır. Etmenin tanısında uygulanan mevcut izolasyon ve teşhis çalışmaları yaklaşık bir hafta sürmekte ve tür bazında teşhisleri zor olmakta, ayrıca patojenite testleri ile bu süre uzamaktadır. Spor yapılarına bakılarak gerçekleştirilen bu teşhis yöntemlerinin çok fazla zaman, emek ve taksonomik uzmanlık gerektirmesi, daha hassas, güvenilir ve hızlı olan PCR tekniklerinin çalışmamızda tercih edilmesine sebep olmuştur. Bu çalışma ile günümüzde birçok alanda yaygın kullanımı olan hem geleneksel PCR hem de Real-Time PCR yöntemi ile Fusarium oxysporum Schltdl izolatının hızlı ve doğru şekilde tespiti gerçekleştirilmiş ve sekanslama. ile yöntemin geçerliliği ortaya konulmuştur.
Turkey has a huge potential in terms of plant genetic resources because it is located in a region that is the cutting point of three major genetic centers in the world. In our country’s species wealth, natural flower beans have a separate place. Although these plants have been used for medical purposes for centuries, because of their flowering in the winter months, they are geneticly used as a decorative plant in gardens. Fusarium plagues, which are commonly found in the plagued decorated plants that contain many pathogens, such as leafy leafy, root fragrance and swelling factors, cause major economic losses every year. The existing isolation and diagnosis studies applied to the diagnosis take about a week and diagnosis on the basis of the species is difficult, and this period is also extended with pathogenity tests. The fact that these methods of diagnosis performed in terms of sports structures require a lot of time, work and taxonomic expertise has led to the preference of PCR techniques that are more accurate, reliable and fast in our work. With this study, both the traditional PCR and Real-Time PCR method, which is widely used in many areas today, has been rapidly and accurately detected and sequenced the Fusarium oxysporum Schltdl isolate. The validity of the method has been revealed.
