Istranca Meşesinin (Quercus hartwissiana Steven) Nozokomiyal Patojenlere Karşı Antimikrobiyal Aktivitesi

Amaç: Son zamanlarda antibiyotiklerin bilinçsiz kullanımı ile patojen mikroorganizmaların direnç kazanması, yeni antibiyotik arayışlarına neden olmaktadır. Bu çalışmada, ıstranca meşesi (Quercus hartwıssıana Steven) kabuğu ekstraktlarının, bakteriyel ve fungal patojenlere karşı antimikrobiyal aktivitesinin araştırılması amaçlanmıştır. Gereç ve Yöntemler:  Istranca meşesinin, etanol, formaldehit, aseton, etil asetat ve metanol çözücüleri ile ekstraksiyonu gerçekleştirilmiştir. Bu ekstraktlardan 50µl alınarak steril disklere emdirildikten sonra ekstraktlarda bulunan çözücülerin sonucu etkilememesi amacıyla diskler 24 saat steril ortamda kurutulmuştur. Disk difüzyon yöntemi ile Pseudomonas aeruginosa, Acinetobacter baumannii, Escherichia coli, Klebsiella pneumoniae, Salmonella spp., metisilin dirençli Staphylococcus aureus (MRSA), metisilin duyarlı Staphylococcus aureus (MSSA), Staphylococcus epidermidis, Enterococcus spp., Candida albicans, Candida glabrata ve Candida tropicalis türlerine karşı antimikrobiyal aktivite araştırılmıştır Bulgular: Çalışmada, etanolle hazırlanan ekstraktıngeniş zon çapını Enterococcus spp.(8 mm) ve S. epidermidis (8 mm) suşlarında; formaldehit ve etil asetatla hazırlanan ekstraktlarıngeniş zon çapını E. coli suşunda (16 mm, 28 mm); asetonla hazırlanan ekstraktıngeniş zon çapını Enterococcus spp. suşunda (21 mm); metanolle hazırlanan ekstraktın isegeniş zon çapını MSSA suşunda (28 mm) oluşturduğu belirlenmiştir. Mayalarda ise, etanolle hazırlanan ekstraktıngeniş zon çapını C. glabrata suşunda (12 mm); formaldehitle hazırlanan ekstraktıngeniş zon çapını C. albicans (28 mm) ve C. tropicalis (28 mm) suşlarında; aseton, etil asetat ve metanolle hazırlanan ekstraktlarıngeniş zon çapını C. albicans suşunda(19 mm, 30 mm, 22 mm) oluşturduğu saptanmıştır. Sonuç:: Istranca meşesinin etil asetat ile elde edilen ekstraktının diğer çözücülerle elde edilen ekstraktlara oranla, çalışmaya alınan suşlarda daha geniş duyarlılık zon çapı oluşturduğu saptanmıştır. Ayrıcageniş zon çapını E.coli, S.aureus ve C.albicans türlerinde oluşturduğundan, ülkemizde doğal olarak yetişen ıstranca meşesinin inhibe edici etkisine yönelik in vivo antimikrobiyal çalışmalar yapılmasının önemli olduğu düşünülmüştür.

Abstract:

Purpose: Recently, the unconscious use of antibiotics and the gain of resistance of pathogenic microorganisms lead to new antibiotic searches. In this study, it was aimed at researching the antimicrobial activity of curl extract (Quercus hartwıssıana Steven) against bacterial and fungal pathogens. Tools and Methods: The extraction of the strawberries is carried out with ethanol, formaldehyde, acetone, ethyl acetate and methanol solvents. After absorbing 50μl of these extracts into the sterile discs, the discs are dried in a sterile environment for 24 hours in order not to affect the result of the solvents found in the extracts. The method of disque diffusion is Pseudomonas aeruginosa, Acinetobacter baumannii, Escherichia coli, Klebsiella pneumoniae, Salmonella spp. Methylococcus aureus (MRSA), methylococcus aureus (MSSA), Staphylococcus epidermidis, Enterococcus spp., Candida albicans, Candida glabrata and Candida tropicalis species have been studied antimicrobial activity: In the study, ethanole prepared the extractive extractive zone diameter Enterococcus spp.(8 mm) and S. epidermidis (8 mm); extracts prepared with formaldehyde and ethyl acetate, extracts prepared with E. coli (16 mm, 28 mm); extracts prepared with acetate, extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts prepared with the extracts. In glabrata swallow (12 mm); the diameter of the extractive extractive zone prepared with formaldehyde was found in C. albicans (28 mm) and C. tropicalis (28 mm) swallow; the diameter of the extractive zone prepared with acetone, ethyl acetate and metanole was found in C. albicans swallow (19 mm, 30 mm, 22 mm). The result: The extract obtained by ethyl acetate is found to be a wider sensitivity zone diameter in the studies obtained in comparison to the extract obtained by other solvent. Since the speculative zone diameter is formed in the E.coli, S.aureus and C.albicans species, it is considered important to conduct in vivo antimicrobial studies on the inhibitory effect of naturally grown strawberries in our country.
