Bu çalışmanın amacı Afrika gri papağanlarında (Psittacus erithacus) cinsiyet tayininde kullanmak üzere dökülmüş tüylerden DNA izolasyonunda iki metodun (metot 1 ve metot 2) etkinliğinin değerlendirilmesidir. Oniki tanesi üç aya kadar saklanmış; iki tanesi dışkıyla kontamine, ayrıca sekiz tanesi beş aya kadar saklanmış; biri dışkıyla kontamine olmak üzere toplam yirmi papağana ait dökülmüş tüy örnekleri kullanılmıştır. Üç aya kadar saklanmış ve dışkıdan ari on adet dökülmüş tüyden metot 1 ile genomik DNA başarı ile elde edilmiştir. Cinsiyet ayırımı kromo helikaz-bağlanma bölgesi genlerinin (CHDW ve CHDZ) çoğaltılması ile iki dişi sekiz erkek olarak sonuçlanmıştır. Ancak genomik DNA metot 1 ile dışkı ile kontamine olmuş tüyler ile birlikte beş aya kadar saklanmış olan tüylerin hiçbirinden, ayrıca metot 2 ile tüy örneklerinin tamamından elde edilememiştir. Dökülmüş tüylerin dışkı ile kontaminasyonu ve tazeliği cinsiyet tayininin başarısını etkilemektedir. Sonuç olarak metot 1, Afrika gri papağanlarında temiz ve taze dökülmüş tüylerden DNA izolasyonu yapılarak cinsiyet tayininde kullanılabilir.
The aim of this study is to evaluate the effectiveness of two methods (methode 1 and method 2) in DNA isolation from scratched hair for gender determination in African gray papaya (Psittacus erithacus). Twelve of them were hidden for up to three months; two of them were contaminated with the output, and eight of them were hidden for up to five months; one was used with a total of twenty papagans, which were contaminated with the output. Up to three months hidden and out of the output, ten pieces of depleted thin meth 1 was obtained with success by genomic DNA. The gender distinction resulted in the reproduction of the chromic helicase-binding zone genes (CHDW and CHDZ) in two female eight men. However, the genomic DNA method 1 was not obtained from any of the hair that was hidden for up to five months along with the hair contaminated with the output, as well as from all the hair samples with the method 2. The pollution and freshness of the output of scratched hair affects the success of the gender determination. As a result, the method 1 can be used in gender determination by making DNA isolation from clean and fresh wrapped hair in African gray papaya.
