Bu çalışma, sevofluran'ın parankim organlar üzerindeki morfolojik değişikliklere likopenin antioksidan etkisinin incelenmesi amacıyla yapıldı. Yirmidört adet albino Wistar rat kontrol, sevoflurane ve likopen olmak üzere üç eşit gruba ayrıldılar. On gün süreyle kontrol ve sevoflurane gruplarındaki ratlara gavajla plasebo (% 0.9 serum fizyolojik), likopen grubundakilere ise gavajla lycomato (Lyc-O-Mato, 40 mg/kg/gün) verildi. Daha sonra likopen ve sevoflurane grupları %50 Oksijen ve %50 N2O karışımı (4L/dk) içinde %3 sevofluran olacak şekilde 2 saat süreyle hazırlanan özel bir düzenek ile uygulandı. Anestezi sonlandırıldıktan hemen sonra bütün ratlar ötenazi edilerek nekropsileri yapıldı. Histopatolojik incelemede ışık mikroskobisi ile karaciğerde konjesyon, sentrolobuler dejenerasyon ve nekroz, böbrekte kortikal nekroz, tubulus epitellerinde dejenerasyon ve nekroz saptandı. Bu değişiklikler sevofluran grubunda şiddetli derecede, likopen grubundaki ratlarda ise daha hafif derecede idi. Histopatolojik skorlamanın istatiksel değerlendirilmesi sonucu sevofluran verilen gruptaki histopatolojik değişimlerin sevofluran ve likopen verilen gruba göre önemli derecede (P≤0.001) daha yaygın olduğu ortaya kondu. Çalışmadan elde edilen sonuçlar likopenin sevoflurane'ın zararlı etkilerini azalttığını göstermektedir.
This study was conducted with the aim of studying the antioxidant effect of sevofluran on the morphological changes on the parankymic organs. Twenty-four albino Wistar rat controls were divided into three equal groups: sevofluran and likopen. For ten days, rats in the control and sevoflurane groups were given plasebo (% 0.9 serum physiological), and likopen groups lycomato (Lyc-O-Mato, 40 mg/kg/day). Later, the likopen and sevoflurane groups were applied with a special arrangement prepared for 2 hours, so that the 50% oxygen and 50% N2O mixture (4L/min) will be 3% sevofluran. Immediately after the anesthesia was terminated, all the rates were overwhelmed and the necropsy was made. In the histopathological study, light microscopy identified conjection in the liver, centrolobular degeneration and necrosis, cortical necrosis in the kidneys, degeneration and necrosis in tubulus epithels. These changes were severe in the sevofluran group, and lighter in the rat in the likopen group. The statistical assessment of the histopathological score found that the histopathological changes in the sevofluran group were significantly more common (P≤0.001) than in the sevofluran and likopen group. Results from the study show that likopenin reduces the harmful effects of sevoflurane.
