Bu çalışmada, ekonomik değeri olan Solanum tuberosum L. cv. Marfona’da in vitro mikrotuber oluşumu üzerinde fotoperiyot ile ortama ilave edilen Jasmonik asit (JA), aktif kömür (AC) ve agarın etkileri araştırılmıştır. Bu amaçla çalışma 4 aşamada gerçekleştirilmiştir: Tuberlerden sürgün oluşumu, in vitro sürgün ucu kültürü, mikroçoğaltım ve mikrotuberizasyon. Sürgün ucu kültürü ile in vitro çoğaltılan bitkilerden alınan tek boğum eksplantları JA (0.0-0.2 mg L-1), agar (%0.5, %0.7) ve AC (0, %0.2) içeren çift fazlı Murashige and Skoog (MS) ortamlarında 2 farklı fotoperiyotta kısa gün (8/16 saat aydınlık/karanlık veya sürekli karanlık) kültüre alınmıştır. En yüksek mikrotuber verimi (%137) ve en yüksek ortalama mikrotuber ağırlığı (183 mg); % 0.2 AC, %0.5 agar içeren çift fazlı MS ortamında ve karanlıkta elde edilmiştir. AC’nin mikrotuber oluşumu üzerindeki olumlu etkisinin JA’den daha fazla olduğu ve AC’nin karanlıkta daha etkili olduğu belirlenmiştir.

In this study, the economic value of Solanum tuberosum L. cv. The effects of Jasmonic acid (JA), active coal (AC) and agarine, added to the environment with photoperyot on the formation of in vitro microtubes in Marfona, have been studied. For this purpose, the work was carried out in 4 stages: the formation of the exile from tubes, the culture of the exile in vitro, the microtuberization and microtuberization. The single bulk explanations taken from in vitro multiplied plants with extractive end culture have been cultivated in 2 different photopery in a double-phase environment in Murashige and Skoog (MS) containing JA (0.0-0.2 mg L-1), agar (0.5, 0.7 %) and AC (0.0.2 %) for a short day (8/16 hours of light/dark or constant darkness). The highest microtuber efficiency (137%) and the highest average microtuber weight (183 mg); 0.2% AC, 0.% AC. The double-phase MS containing 5 agar is obtained in the environment and in the dark. The positive effect of AC on the formation of microtubes has been determined to be greater than JA and AC is more effective in darkness.
