Amaç: Bu çalışmada kültür vasatına ilave edilen L-Ergotiyonin ve Fetuin?in blastosist gelişimine etkisinin belirlenmesi amaçlanmıştır. Gereç ve Yöntem: Çalışmanın materyalini mezbahadan alınan ovaryumlardan toplanan oositler oluşturmuştur. Maturasyon (IVM) medyumunda 22 saat inkübe edilen oositler fertilizasyon (IVF) medyumunda sperma ile 20-22 saat inkübe edilmiştir. Fertilizasyon aşamasından sonra antioksidan (Grup 1: L-Ergotiyonin (L-Erg) 0.05 mM (n:100), Grup 2: Fetuin 1 mg/ml (n:100), Grup 3: kontrol (n:120) ilave edilen kültür (IVC) droplarına aktarılan zigotlar kültüre edilmiştir. Blastosist gelişimi 6. ve 7. günde kontrol edilmiştir. İn vitro embriyo üretim aşamalarında oluşan farklılıklar Ki-kare testi ile değerlendirilmiştir. Bulgular: In vitro maturasyona alınan toplam 320 oositin 299?unun (%93,44) mature olduğu belirlenmiştir. Grup I, II ve III?ün maturasyon oranları sırasıyla %94, %92 ve %94,17 olarak tespit edilmiştir. Grupların cleavage oranlarının sırasıyla %92,55, %94,57 ve %92.04 olduğu belirlenmiştir. Kültür aşamasına alınan 299 zigottan 65 (%21,74) adet blastosist elde edilmiş ve gruplardaki blastosist oranlarının sırasıyla %45,74, %11,96 ve %18,58 olduğu görülmüştür. Yapılan çalışma sonrasında blastosist oranlarında L-Erg ve diğer iki grup arasında istatistiki farkın önemli olduğu belirlenmiştir (p<0.05). Öneri: Sonuç olarak L-Erg?in blastosist gelişim oranlarını iyileştirdiği görülmüştür. L-Erg?in farklı dozlarda kullanımının araştırılarak antioksidan etkinliğinin en yüksek olduğu dozunun in vitro embriyo üretiminde kullanılabileceği ve daha fazla sayıda blastosist üretilebileceği düşünülmüştür.
Purpose: This study is aimed at determining the effect of L-Ergotiion and Fetuin on blastosistic development added to the culture. Tool and Method: The material of the study was formed by oocytes collected from the ovaries taken from the mosquito. Oocytes incubated in the maturing (IVM) medium for 22 hours are incubated with sperm in the fertilization (IVF) medium for 20-22 hours. After the fertilization phase, zigots transmitted to the antioxidants (Group 1: L-Ergotionin (L-Erg) 0.05 mM (n:100), Group 2: Fetuin 1 mg/ml (n:100), Group 3: Control (n:120) added to the culture (IVC) drops were cultivated. The development of blastosist was controlled on the 6th and 7th days. The differences in the embryos production stages in vitro have been assessed by Ki-core test. Results: In vitro maturity is estimated that the total of 320 oosithins received at 299 (93.44) are mature. Groups I, II and III’s maturity rates were 94, 92% and 94,17% respectively. The cleavage rates of the groups were 92.55% respectively, 94.57% and 92.04%. Of the 299 zigots taken to the cultural stage, 65 (21.74) blastosists were obtained and the blastosist rates in the groups were 45.74, 11.96 and 18.58 respectively. After the study was determined that the statistical difference between L-Erg and the other two groups in blastosist rates was significant (p<0.05). Recommendation: As a result, it has been shown that L-Erg improves the blastosist development rates. The use of L-Erg in different doses has been studied and it is believed that the dose with the highest antioxidant effectiveness can be used in in vitro embryonic production and more blastosists can be produced.
