Amaç: Bu çalışmada, fare L929 fibroblast hücrelerinde H2O2kaynaklı oksidatif stresi önlemede ve yara iyileşme modelinde Gossypin’in kapasitesini değerlendirdik. Materyal ve Metot: Gossypin’in (5–200 µg/mL) ve H2O2’nin (0,3–0,9mM) in-vitro yara iyileşme aktivitesi, hücre canlılığı ve sitotoksisite indeksleri MTT tahlili kullanılarak fare fibroblast hücre hattı L929’da değerlendirildi. Çalışmada, in-vitro yara iyileşme testi modeli olarak migrasyon testi yapıldı. Antioksidan aktivitenin değerlendirilmesi amacı ile H2O2’nin (0,5 mM) konsantrasyonu kullanıldı. SOD aktivitesi ve MDA seviyesi belirlendi. Ayrıca Gossypin dozlarının H2O2’nin neden olduğu hasarı azaltıp azaltamadığını değerlendirmek için, gerçek zamanlı PCR kullanarak L929 hücrelerinde IL-1β ve TNF-α mRNA ekspresyon seviyelerini analiz ettik. Bulgular: H2O2 (0,3–0,9 mM) uygulamasının doz bağımlı olarak, Gossypin’in (5–200 µg/mL) ise 75 µg/mL’den sonra L929 hücrelerine proliferasyonu inhibe ettiği belirlenmiştir. Gossypin’in 25 ve 50 µg/mL dozları H2O2 (0,5 mM) uygulamasına karşı 48. ve 72. saatlerde L929 hücrelerini korumuştur. Bu sonuçlar migrasyon testi ile doğrulanmıştır. Gossypin uygulanması, antioksidan aktivitesini önemli ölçüde arttırmış, MDA oluşumunu azaltmıştır. Özellikle 50 μg/mL grubu ve H2O2 grubuna kıyasla SOD’u arttırmıştır. Gossypin dozları değerlendirilen H2O2 grubuna kıyasla IL-1β ve TNF-α mRNA ekspresyonunu anlamlı şekilde azaltmıştır. Sonuç: Bu çalışmada, L929 hücrelerinde H2O2 tarafından indüklenen oksidatif stresinden koruyarak fibroblast canlılığı ve migrasyonu üzerinde güçlü bir etki göstermiştir. Bu, Gossypin’in H2O2’nin neden olduğu oksidatif stres ile ilişkili hastalıklarının önlenmesinde potansiyel etkisini ortaya koymaktadır. Buda cilt dokusunun H2O2’nin neden olduğu oksidatif stres benzeri hasarlanmalara bağlı hastalıkların tedavisinde Gossypin’in kullanılabileceğini göstermektedir.
Purpose: In this study, we evaluated the capacity of Gossypin in the prevention of H2O2 source oxidative stress in mouse L929 fibroblast cells and in the wound healing model. Material and Method: Gossypin (5-200 μg/mL) and H2O2 (0,3-0,9mM) in-vitro wound healing activity, cell vitality and cytotoxicity indices were evaluated in the mouse fibroblast cell line L929 using MTT assessment. In the study, a migration test was performed as a in-vitro wound healing test model. The antioxidant activity was evaluated with the H2O2 (0,5 mM) concentration. SOD activity and MDA levels have been determined. In addition, to assess whether doses of Gossypin can reduce the damage caused by H2O2, we analyzed the levels of expression of IL-1β and TNF-α mRNA in L929 cells using real-time PCR. Results: H2O2 (0,3-0,9 mM) is determined depending on dosage, while Gossypin (5-200 μg/mL) inhibits proliferation to L929 cells after 75 μg/mL. Gossypin’s 25 and 50 μg/mL doses protect the L929 cells in 48 and 72 hours against the application of H2O2 (0,5 mM). These results are confirmed by a migration test. The application of Gossypin has significantly increased its antioxidant activity, reduced the formation of MDA. It has increased SOD in particular compared to the 50 μg/mL group and the H2O2 group. Gossypin doses significantly reduced the expression of IL-1β and TNF-α mRNA compared to the assessed H2O2 group. Result: In this study, the L929 cells have shown a strong effect on fibroblast vitality and migration by protecting them from oxidative stress induced by H2O2. This shows the potential effect of Gossypin in the prevention of diseases associated with oxidative stress caused by H2O2. Buda shows that Gossypin can be used in the treatment of diseases associated with oxidative stress-like damage caused by H2O2 in the skin tissue.
