Bu çalışmada; gıda katkı maddesi Tartrazin’in Allium cepa L. hücrelerinde meydana getirdiği fizyolojik, anatomik ve sitogenetik etkiler araştırılmıştır. Fizyolojik parametreler olarak çimlenme yüzdesi, kök uzunluğu ve ağırlık artışı; sitogenetik parametreler olarak kromozom hasarlar, mikronukleus (MN) ve mitotik indeks (MI) sıklığı; anatomik parametreler olarak ise anatomik hasarlar incelenmiştir. Uygulama grubundaki tohumlar 50, 100 ve 200 ppm dozunda Tartrazin ile 72 saat süresince muamele edilmiş ve elde edilen veriler istatistiksel olarak değerlendirilmiştir. Sonuç olarak, kontrol grubuyla karşılaştırıldığında çimlenme yüzdesi, kök uzunluğu, ağırlık artışı, kromozom anormallikler, MI ve MN sıklığı ile kök anatomisinde önemli değişimler saptanmıştır. Tartrazin’in tüm uygulama gruplarında doza bağlı olarak çimlenme yüzdesi, kök uzunluğu, ağırlık artışı ve MI’i azaltığı; kromozom anormallikleri, MN sayısı ve anatomik hasarları ise arttırdığı tespit ediliştir. Yapılan mikroskobik incelemeler sonucunda, Tartrazin uygulamasının A. cepa L. kök ucu hücrelerinde fragment, yapışkan kromozom, kromozom köprüsü, kromatinin eşit olmayan dağılımı ve C- mitoz şeklinde kromozomal hasarların oluşumunu teşvik ettiği görülmüştür. Ayrıca, Tartrazin uygulaması kök ucu hücrelerinde yassılaşmış hücre çekirdeği, korteks hücrelerinde bazı maddelerin birikimi, hücre deformasyonu, belirgin olmayan iletim doku, nekroz ve korteks hücre çeperlerinde kalınlaşma şeklinde anatomik hasarların oluşumuna da sebep olmuştur.
In this study, the physiological, anatomical and cytogenetic effects of food additives Tartrazin occurred in the cells of Allium cepa L. As physiological parameters, grinding percentage, root length and weight increase; chromosome damage as cytogenetic parameters, micronucleus (MN) and mitotic index (MI) frequency; and anatomical damage as anatomical parameters. The seeds in the application group were treated with Tartrazin in doses of 50, 100 and 200 ppm for 72 hours and the data obtained were statistically evaluated. As a result, compared to the control group, significant changes in the root anatomy have been detected with grinding percentage, root length, weight increase, chromosome abnormalities, MI and MN frequency. It has been found that Tartrazin in all the application groups has reduced the grinding percentage, root length, weight increase and MI depending on the dose, while the chromosome anomalies, the number of MN and anatomical damage are increased. Microscopic studies have shown that the application of Tartrazin in the A. cell L. root end cells promotes fragment, adhesive chromosome, chromosome bridge, uneven distribution of chromatine and the formation of chromosomal damage in the form of C-mitoz. Furthermore, the application of Tartrazin has also caused the formation of anatomical damage in the form of thickness in the root end cells, the accumulation of certain substances in the cortex cells, cell deformation, unseen transmission tissue, necrosis and cortex cell circles.
