Erişkin erkek sıçanlarda karbon tetraklorür ile oluşturulan testis hasarı üzerine kuersetinin etkisi / Effect of quercetin on CCl₄ induced testicular injury in adult male rats

Özet: Karbon tetraklorür (CCl₄), endüstride soğutucu floroklorokarbonların üretiminde kullanılan organik bir bileşiktir. CCl₄ testiküler hücrelere hasar vererek erkek üreme fonksiyonlarını olumsuz yönde etkiler. Kuersetin, antioksidan özellikleri iyi bilinen bir flavonoiddir. Bu çalışmada CCl₄ uygulanmış erkek sıçanların testisleri üzerinde CCl₄’ün oluşturduğu hasarda kuersetinin etkisini incelemeyi amaçladık. Bu çalışmada toplam 28 adet erişkin Spraque-Dawley türü erkek sıçan kullanıldı. Deney hayvanları; Kontrol grubu, CCl₄ grubu, kuersetin, CCl₄ + kuersetin grubu olmak üzere toplam 4 gruba ayrıldı. Bu deney hayvanlarından; kontrol grubuna 7 gün boyunca günde bir kez belirlenen dozda zeytinyağı, CCl₄ grubuna 7 gün boyunca günde bir 0.5 ml/kg CCl₄, kuersetin grubuna 7 gün boyunca günde bir tek dozda 20 mg/kg kuersetin, CCl₄ + kuersetin grubuna ise 7 gün boyunca günde bir kez 0.5 ml/kg CCl₄ ve 7 gün boyunca günde bir 20 mg/kg kuersetin intraperitoneal yolla uygulandı. Deney hayvanları deney süresi sonunda anestezik madde ile uyutuldu ve testis dokusu örnekleri Bouin solüsyonu içerisine alındı. Örnekler rutin histolojik işlemlerin ardından bloklandı. Her bir bloktan 3 μm kesit alınarak Hematoksilen-Eozin ve Periyodik Asit-Schiff + Hematoksilen boyalarıyla boyandı ve mikroskobik incelemeleri yapıldı. Çalışma sonucunda elde edilen bulgulara göre, kontrol grubundaki sıçanların testislerinde normal histolojik yapı, CCl₄ uygulanmış sıçanların testislerinde yoğun spermatogenetik hücre hasarı, CCl₄ + kuersetin verilen grupta ise azalmış hücre hasarı gözlendi. CCl₄’ün erkek sıçanların testisleri üzerinde toksisiteye yol açtığı, kuersetinin ise CCl₄’ün testislerde oluşturduğu bu hasarı azalttığı görüldü. In this study, the testes of male rats on applied CCl₄ damage caused by CCl₄ aimed to investigate the effect of quercetin. In this study, a total of 28 adult male Spraque Dawley rats were used. Laboratory animals; Control group, CCl₄ group, quercetin group, including CCl₄ + quercetin group was divided into 4 groups. This experimental animals; control group for 7 days, once daily determined dose of olive oil, CCl₄ group daily for 7 days of a 0.5 ml/kg CCl₄, quercetin group daily for 7 days in a single dose of 20 mg/kg of quercetin, CCl₄ + quercetin group for 7 days once a day 0.5 ml/kg daily for 7 days CCl₄ 20 mg/kg were injected with quercetin. Experimental animals were anesthetized with anesthetic agent at the end of test period, and testis tissue samples taken up in Bouin solution. After the staining process was blocked. 3 mm sections taken from each block hematoxylin and eosin and periodic acid-Schiff + were stained with hematoxylin and microscopic examinations were performed. According to the findings obtained from the study, the normal histological structure of the testis of rats in the control group, intense spermatogenic cells in the testes of rats by CCl₄ injury, CCl₄ + quercetin decreased cell damage was observed in the group. As a result, the testes of male rats on CCl₄-induced toxicity, quercetin is formed in the testes of CCl₄ was found to reduce this damage. Keywords: carbon tetrachlorid, quercetin, rat, testes

Carbon tetracloride (CCl4) is an organic compound used in the production of refrigerant fluoroclorocarbons in the industry. CCl4 affects male reproductive functions negatively by damaging testicular cells. Curcetin is a well-known flavonoid with its antioxidant properties. In this study, we aimed at studying the impact of CCl4 on the damage caused by CCl4 on the testicles of male rats applied CCl4. In this study, a total of 28 adult Spraque-Dawley species male rats were used. The experimental animals were divided into a total of 4 groups, including Control Group, CCl4 Group, Circetin, CCl4 + Circetin Group. From this experimental animal; to the control group, the dose of olive oil once a day for 7 days, to the CCl4 group for 7 days is 0. 5 ml/kg CCl4 was administered in a single dose of 20 mg/kg per day for 7 days, and CCl4+ was administered in a single dose of 0.5 ml/kg CCl4 per day for 7 days, and in a single dose of 20 mg/kg per day for 7 days. The experimental animals were dropped with an anesthetic at the end of the experimental period and testis tissue samples were taken into the Bouin solution. Examples were blocked after routine histological processes. 3 μm cut from each block was painted with Hematoxylene-Eosine and Periodic Acid-Schiff + Hematoxylene paintings and microscopic examinations were carried out. According to the findings obtained in the study, normal histological structure in the testicles of the rats in the control group, intense spermogenetic cell damage in the testicles of the CCl4 applied rats, and reduced cell damage in the given group of CCl4 + curcetin. It has been found that CCl4 causes toxicity on the testicles of male rats, while the circle reduces this damage that CCl4 causes in the testicles.
