Amaç: Bu çalışma Çanakkale ve Tekirdağ illeri kanola üretim alanlarındaki bazı virüs hastalıklarının tespiti ve moleküler karakterizasyonu amacı ile yürütülmüştür. Materyal ve Metot: Kanola bitkileri görsel olarak incelenmiş, virüs ve virüs-benzeri simptom gösteren bitkilerden örnekler toplanmıştır. Toplanan örnekler turnip mosaic virus (TuMV), cucumber mosaic virus (CMV) ve cauliflower mosaic virus (CaMV)'ün varlığının belirlenmesi amacı ile DAS-ELISA ile testlenmiştir. Enfekteli bulunanlar içerisinden her bir virüs için 2 izolat seçilerek RT-PCR ile istenilen gen bölgeleri çoğaltılmış, klonlanmış ve sekanslanmıştır. Elde edilen sekans dizileri kullanılarak çoklu dizi ve filogenetik analizler gerçekleştirilmiştir. Bulgular: Testlemeler sonucunda 16 örnek TuMV ile 3 örnek ise CMV ile enfekteli olarak bulunmuştur. Bir örnekte ise CMV ve TuMV karışık enfeksiyonu tespit edilmiştir. Toplanan örneklerin hiçbirisinde CaMV enfeksiyonuna rastlanılmamıştır. İzolatların moleküler karakterizasyonları sonucunda TuMV izolatlarının dünya izolatları ile %78-94 ve 88-98, CMV izolatlarının ise %75-98 ve 80-100 oranında sırası ile nt ve aa düzeyinde benzerlikler gösterdiği tespit edilmiştir. Filogenetik analizler sonucunda ise TuMV izolatlarının Asian BR ve CMV izolatlarının ise IA gruplarında yer aldığı belirlenmiştir. Sonuç: Gerçekleştirilen bu çalışma ile kanola üretim alanlarında TuMV ve CMV enfeksiyonu tespit edilerek, moleküler olarak karakterize edilmiştir.
Purpose: This study was carried out with the aim of detecting and molecular characterization of some viral diseases in the canola production fields of Chanakkale and Tekirdağ. Material and Method: Canola plants are visually studied, samples of plants showing viral and viral-like symptoms are collected. The samples collected were tested with DAS-ELISA to determine the presence of turnip mosaic virus (TuMV), cucumber mosaic virus (CMV) and cauliflower mosaic virus (CaMV). For each virus, 2 isolates are selected and the genes required are multiplied, cloned and scansed with RT-PCR. Multiple series and philogenetic analyses were performed using the sequence series obtained. Results: 16 examples of TuMV and 3 examples of CMV were found infected. In one example, a mixed infection of CMV and TuMV has been detected. None of the samples collected found CaMV infection. The molecular characteristics of the isolat have found that the isolates of TuMV are similar to the world isolates 78-94 and 88-98, and the isolates of CMV are similar to the levels of nt and aa at 75-98 and 80-100 respectively. The philogenetic analysis has found that TuMV isols are in Asian BR and CMV isols in the IA groups. The result: This study was carried out by the detection of TuMV and CMV infections in canola production areas, characterized as molecular.
