Metotreksat ile Karaciğer Hasarı Oluşturulmuş Sıçanlarda Silymarin‘in Koruyucu Etkilerinin Araştırılması

Abstract:

Amaç: Metotreksat (MTX)'ın sıçan karaciğer dokusunda meydana getirdiği hasara karşı silymarin (SİL)'in koruyucu etkilerinin incelenmesi amaçlanmıştır. Gereç ve Yöntem: Çalışmada 36 adet erişkin Sprague Dawley tipi erkek sıçan kullanıldı. Deney hayvanları her grupta 6 hayvan olacak şekilde 6 gruba ayrıldı. 5 günlük deney süresince Grup 1'e 0.5 mL intraperitoneal (i.p) tek doz serum fizyolojik, Grup 2'ye karboksi metil selüloz (CMC) oral olarak, Grup 3'e CMC içinde çözdürülmüş 300 mg/kg Silymarin deney süresince oral yolla, Grup 4'e tek doz 20 mg/kg (i.p) MTX(i.p), Grup 5'e tek doz 20 mg/kg MTX (i.p) + oral CMC, Grup 6'ya ise tek doz 20 mg/kg MTX (i.p) + CMC içinde çözdürülmüş 300 mg/kg dozunda SİL deney süresince oral olarak uygulandı. Deney sonunda sıçanlar dekapite edilerek karaciğer dokuları çıkartıldı. Rutin doku takip işlemlerinden sonra değerlendirildi. Bulgular: Grup 1, Grup 2 ve Grup 3'e ait karaciğer dokuları histopatolojik ve immunohistokimyasal olarak incelendiğinde birbirlerine benzer idi. Grup 4 ve 5'de histopatolojik bulgular benzer olup vasküler konjesyon, sinüzoidal dilatasyon ve inflamatuar hücre artışı görüldü. Yine bu gruplarda TUNEL, Bax ve Kaspaz 3 pozitif hücreler diğer deney gruplarına göre artmış olarak tespit edildi. Grup 6 de ise doku hasarının önemli ölçüde azaldığı gözlendi. Sonuç: MTX karaciğerde önemli doku hasarına neden olmuş ve silimarin bu hasarı iyileştirmiştir.

Abstract:

Purpose: Methotrexate (MTX) is intended to study the protective effects of silymarin (SIL) against the damage caused by methotrexate (MTX) in the mushroom tissue. In the study, 36 adult Sprague Dawley-type male rats were used. The experiments were divided into 6 groups in 6 animals in each group. During the 5 days of the trial, a single dose of 0.5 mL intraperitoneal (i.p) serum was physiological, a single dose of carboxyl methyl cellulose (CMC) was oral, a single dose of 300 mg/kg of silymarin solved in CMC, a single dose of 20 mg/kg (i.p) MTX(i.p), a single dose of 20 mg/kg MTX (i.p) + oral CMC, a single dose of 20 mg/kg MTX (i.p) + CMC solved in CMC, a single dose of 300 mg/kg of silymarin solved in CMC, a single dose of 20 mg/kg MTX (i.p) + oral CMC, and a single dose of 20 mg/kg MTX (i.p) + CMC solved in CMC. At the end of the test, the mice was decaptivated and the liver tissue was removed. The routine was evaluated after the tracking processes. Results: The liver tissues of Group 1, Group 2 and Group 3 were similar to each other when studied histopathological and immunohistochemical. In Groups 4 and 5, histopathological findings were similar, and vascular conjunction, sinusoidal dilatation and inflammatory cell increase were observed. Again, the positive cells of TUNEL, Bax and Kaspaz 3 in these groups were found to have increased compared to other experimental groups. The group 6 observed that tissue damage was significantly reduced. The result: MTX has caused substantial tissue damage in the liver and silimarine has healed this damage.
