Rheum ribes Özütlerinin Antioksidan ve Antibakteriyel Aktiviteleri

Polygonaceae familyasının türü olan Rheum ribes’in taze gövde ve yaprakları Lübnan, İran ve Türkiye’nin doğusunda sebze olarak tüketilmektedir. Son zamanlarda farmakolojik alanlarda kullanılan bu bitkiden Asya ülkelerinde saf ilaç materyalleri üretilmektedir. Çalışmamızda, Rheum ribes gövde kısımlarından etil asetat, hekzan, metanol ve saf su çözücüleri kullanılarak 10-100 mg/mL konsantrasyonlarda hazırlanan özütlerin in vitro fitokimyasal, antioksidan ve antibakteriyel aktiviteleri incelenmiştir. Çözücülere bağlı olarak ekstraksiyon verimi değişmekle birlikte en yüksek ekstraksiyon verimi (%25.3), en yüksek total fenolik (0.80±0.00 mg GAE/g) ve flavonoid içerik (0.321±0.00 mg RE/g) 100 mg/mL konsantrasyonda metanol özütünde belirlenmiştir. Rheum ribes metanol gövde özütünün DPPH· süpürme aktivitesi %91.10±0.51 olarak kaydedilmiş olup IC50 dozu 10.50 mg/mL’dir. En yüksek Fe(II) iyonlarını şelatlama aktivitesi %49.70±3.10, indirgeme kapasitesi (0.19±0.00) ve süperoksit anyonu giderme aktivitesi (%39.20±2.40) 100 mg/mL konsantrasyonda metanol özütünde belirlenmiştir. Rheum ribes’in etil asetat, hekzan ve metanol özütlerinin test bakterileri üzerinde antibakteriyel aktivite gösteremediği belirlenmiştir. Saf su özütünün gösterdiği antibakteriyel aktivite 9.00±1.00 ve 14.50±1.50 mm zon çapı aralığındadır. En yüksek inhibitör etki 14.50±1.50 mm zon çapı ile S. aureus’a karşı gözlenmiştir.

The fresh body and leaves of the Rheum ribes, a species of the Polygonaceae family, are consumed as vegetables in Lebanon, Iran and eastern Turkey. This plant, which is recently used in pharmacological fields, produces pure medicinal materials in Asian countries. In our study, the phytochemical, antioxidant and antibacterial activities of the extract prepared in concentrations of 10-100 mg/mL using ethyl acetate, hexane, methanol and pure water solutions from the body parts of the Rheum ribes were studied in vitro. While the extraction efficiency varies depending on the solvents, the highest extraction efficiency (25.3%), the highest total phenolic (0.80±0.00 mg GAE/g) and flavonoid content (0.321±0.00 mg RE/g) are determined in the 100 mg/mL concentration of methanol extract. Rheum ribes metanol body extract's DPPH· spray activity was recorded at 91.10 ± 0.51%, and the IC50 dose is 10.50 mg/mL. The highest Fe(II) ions are shelating activity of 49.70 ± 3.10, reduction capacity (0.19 ± 0.00) and superoxid anion elimination activity (39.20 ± 2.40%) in the 100 mg/mL concentration of methanol extract. Rheum ribes have been found that ethyl acetate, hexane and methanol extracts do not show antibacterial activity on the test bacteria. The antibacterial activity shown by the pure water extract is in the zone diameter range of 9.00±1.00 and 14.50±1.50 mm. The highest inhibitor effect was observed against S. aureus with a zone diameter of 14.50±1.50 mm.

Recently, crude drug materials from this plant are used in pharmacological research in Asian countries. In this study, in vitro phytochemical, antioxidant and antibacterial activity of the extracts prepared at 10-100 mg/mL concentrations by using ethyl acetate, hexane, methanol and distilled water from stem parts of Rheum ribes were examined. The extraction efficiency varies depending on the solvents and the highest extraction efficiency was calculated as %25.3 at methanol extract. The highest total phenolic and flavonoid contents were determined 0.80 ± 0.00 mg GAE/g and 0.321 ± 0.00 mg RE/g at 100 mg/mL concentration of methanolic extract. DPPH· radical scavenging activity of methanolic stem extract was %91.10±0.51 (IC50 dose 10.50 mg/mL). The highest Fe(II) ions chelating activity (49.70±3.10%), reduction capacity (0.19±0.00) and superoxide anion scavenging activity (39.20 ± 2.40%) were recorded at methanol extract. Ethyl acetate, hexane and methanol extracts of Rheum ribes not showed antibacterial activity against test bacteria. Antibacterial activity of distilled water extract was in the range of 9.00±1.00 and 14.50±1.50 mm. The highest inhibitory effect was observed against S. aureus with 14.5 ±1.50 mm zone diameter.
