Bazı Sertifikalı Armut (Pyrus communis L.) Genotiplerinin Morfolojik ve RAPD Yöntemleriyle Tanımlanması

Abstract:

Bu çalışmada ülkemizde yoğun olarak yetiştiriciliği yapılan 6 armut genotipinde RAPD (Randomly Amplified Polymorphic DNA) tekniği kullanılarak akrabalık ilişkileri belirlenmiştir. Yapılan moleküler analizler sonucunda 8 adet RAPD primeri, 48 adet polimorfik bant üretmiştir. En fazla sayıda polimorfik bant (11 adet), OPA-08 primerinden elde edilirken, en az sayıda polimorfik bant (2 adet) OPE-07 primerinden elde edilmiştir. Primer başına düşen ortalama polimorfik bant sayısı 6 adet olmuştur. Tüm çeşitler arasında, en yüksek benzerlik indeksi Klasik Akça ve Kokulu Akça arasında (0,922) görülürken, en düşük indeks Kokulu Akça ile Santa Maria çeşidi arasında (0,431) tespit edilmiştir. Dendrogramda tüm genotipler 2 ana gruba ve bu ana gruplar da kendi içerisinde 2 alt gruba ayrılmıştır. Klasik Akça ve Kokulu Akça genotipleri birinci alt grupta yer alırken; Deveci armudu ikinci ve Saman Akçası ile Dikenli Akça ise üçüncü alt grupta yer almıştır. Santa Maria çeşidi de dördüncü alt grupta bulunmaktadır. Ayrıca UPOV kriterlerine göre pomolojik analizler de yapılmış olup bu analizler sonucunda genotiplerde meyve ağırlığı (30,19-305,0), meyve eni (3,76-8,15), meyve boyu (5,89-10,6), meyve şekil indeksi (0,94-1,57), meyve eti sertliği (42,03-66,1), meyve kabuk rengi (L 64,11-75,81, a 14,55-6,86, b 27,05-50,3), meyve suyu pH’ı (2,51-5,16), SÇKM (8,95-10,4), meyve sapı uzunluğu (4,95-2,32), meyve sapı kalınlığı (2,8-4,67), sap çukuru eni (3,65-13,09), sap çukuru derinliği (1,03-4,65), çiçek çukuru eni (7,93-13,94), çiçek çukuru derinliği (3,65-10,74), çekirdek evi boyu (23,86-34,73), çekirdek evi eni (11,59-26,98), çekirdek boyu (8,34-9,19), çekirdek eni (2,8-4,68) ve çekirdek kalınlığı (1,1-3,18) bakımından genotipler arasında önemli farklılıklar bulunmuştur.

Abstract:

In this study in our country intensely cultivated 6 genotypes were identified by using the RAPD (Randomly Amplified Polymorphic DNA) technique. The molecular analysis resulted in 8 RAPD primers and 48 polymorphic bands. The maximum number of polymorphic bands (11 pieces) is obtained from the OPA-08 primary, while the minimum number of polymorphic bands (2 pieces) is obtained from the OPE-07 primary. The average number of polymorphic bands per primary was 6. Among all varieties, the highest similarity index is seen between the Classical Flow and the Painful Flow (0,922) while the lowest index is found between the Painful Flow and the Santa Maria varieties (0,431). In the Dendrogram, all genotypes are divided into 2 main groups and these main groups are also divided into 2 subgroups within themselves. Classical Akça and Aroma Akça genotypes are in the first subgroup; Deveci armudu is in the second and Saman Akça and Dikenli Akça is in the third subgroup. The Santa Maria varieties are also in the fourth subgroup. According to UPOV criteria, pomological analyses have also been carried out and these analyses have resulted in the genotypes of fruit weight (30,19-305,0), fruit thickness (3,76-8,15), fruit length (5,89-10,6), fruit shape index (0,94-1,57), fruit meat hardness (42,03-66,1), fruit shape color (L 64,11-75,81, a 14,55-6,86, b 27,05-50,3), fruit juice pH (2,51-5,16), fruit shape length (8,95-10,4), fruit shape thickness (4,95-2,32), fruit shape thickness (2,8-4,67), fruit shape thickness (3,65-13,09), fruit shape thickness (1,03-4,65), fruit shape thickness (L 64,11-75,81, a 14,55-6, b 27,05-50,3), fruit
