Amaç: Bu çalışmada doksorubisin kaynaklı böbrek hasarında Vitamin D'nin iyileştirici etkileri ve asprosin ile ilişkisinin araştırılması amaçlandı. Gereç ve Yöntem: Erkek Wistar Albino sıçanlar her grupta 6 hayvan olacak şekilde 4 gruba ayrıldı. Kontrol grubuna herhangi bir işlem uygulanmadı. Vitamin D grubundaki sıçanlara oral olarak 200 IU/gün D Vitamini verildi. Doksorubisin grubundaki sıçanlara deneyin ilk gününde intraperitoneal (i.p.) olarak tek doz doksorubisin 10 mg/kg oranında uygulandı. Doksorubisin+D vitamini grubundaki sıçanlara tek doz doksorubisin i.p. verildi. Deneyin ilk gününde 10 mg/kg ve 14 günlük deney periyodu boyunca her gün oral olarak 200 IU/gün D vitamini verildi. Çalışma sonunda tüm gruplardaki sıçanlar dekapite edilerek doku ve kan örnekleri alındı. Toplam oksidan seviyesi, kan örneklerinden ELISA yöntemi ile ölçüldü. Böbrek dokuları TUNEL ve immünhistokimyasal boyama ile incelendi. Bulgular: Bu çalışmada doksorubisinin toplam oksidan düzeylerini ve TUNEL pozitifliğini artırdığı, asprosin düzeylerini düşürdüğü, tedavi olarak verilen D vitamininin toplam oksidan düzeyini ve TUNEL pozitifliğini azalttığı, asprosin düzeyini değiştirmediği belirlendi. Sonuç: Doksorubisin'in asprosini azaltarak doku hasar mekanizmalarında etkili olabileceği ve tedavide D vitamininin iyileştirici etkisinin olduğu ancak bu iyileştirici etkinin asprosinden bağımsız bir mekanizma ile sağlandığı gösterilmiştir.
Purpose: This study was aimed at investigating the healing effects of vitamin D on kidney damage caused by dosorubin and the relationship with asprosin. Man Wistar Albino rats were divided into 4 groups so that they will be 6 animals in each group. No action was carried out on the control group. The rats in the D-vitamin group were given 200 IU/day vitamin D orally. The first day of the trial on rats in the Doksorubisin group is intraperitoneal (i.p.) A single dose of doxorubin was administered at a rate of 10 mg/kg. Doksorubin + vitamin D for mice. was given. On the first day of the test, 10 mg/kg and 200 IU/day of vitamin D were given orally every day during the 14 day of the test period. At the end of the study, the mushrooms in all groups were decaptivated and weeds and blood samples were taken. The total oxygen level was measured by the ELISA method from blood samples. The kidney tissues were studied with TUNEL and immune chemical painting. The findings: In this study, it was found that the total oxidate levels and TUNEL positive levels were increased, that the levels of asprosin were reduced, that the total oxidate levels and TUNEL positive levels were reduced, that the levels of asprosin were not changed. The result: Doksorubicin has been shown to be effective in tissue damage mechanisms by reducing asprosin, and vitamin D has a healing effect in treatment but this healing effect is provided by a mechanism independent of asprosin.
