Bu çalışma 2013-2014 yıllarında Denizli ilinde Bekilli (Merkez), Buldan (Çamköy), Çal (Kabalar), Güney (Aydoğdu, Eziler ve Adıgüzel) ve Merkez (Gözler) ilçeleri ile Manisa ilinin Salihli (Poyrazdamları ve Yeşilova) ilçesinde yapılmıştır. Sürvey çalışmaları Denizli ve Manisa illerinde toplam 29 adet kekik tarlasında gerçekleştirilmiştir. Belirlenen tarlalarda köşegenler doğrultusunda dolaşılarak bitkiler hasta-sağlam olarak sayılmış ve hastalık belirtileri kaydedilerek örneklemeler yapılmıştır. Ayrıca il ve ilçe düzeyinde hastalığın yaygınlık oranı belirlenmiştir. Kök ve kök boğazı çürüklüğü belirtisi gösteren bitkilerden toplam 50 izolat elde edilmiştir. Bu izolatlar mikroskop altında morfolojik yapılarına göre Aspergillus niger Tiegh., Alternaria spp., Rhizoctonia solani J.G.Kühn ve Macrophomina phaseolina (Tassi) Goid. olarak tanımlanmıştır. Ön patojenisite testlerinin sonucunda Alternaria spp. ve Aspergillus niger izolatları, bitki parçalarında kahverengileşme ve çürüklük belirtisine neden olmamış ve bu nedenle saksı denemelerine alınmamıştır. Macrophomina phaseolina ve Rhizoctonia solani’ye ait izolatların tamamı saksı denemeleri ile gerçekleştirilen testlerle de desteklenerek kekik bitkisinde patojen olarak belirlenmiştir.
This study was conducted in the years 2013-2014 in the province of Denizli in Bekilli (Central), Buldan (Chamköy), Çal (Kabalar), South (Aydoğdu, Eziler and Adıgüzel) and Central (Gözler) districts and in the province of Manisa in Salihli (Poyrazdamları and Yeşilova). Survey work was carried out in the provinces of Denizli and Manisa in a total of 29 square fields. In the fields specified, the plants were considered patient-healthy and the symptoms of the disease were recorded and samples were made. The prevalence of the disease has also been determined at the province and district level. A total of 50 insulates were obtained from the plants that indicate root and root throat crust. These isolates under the microscope according to the morphological structures of Aspergillus niger Tiegh., Alternaria spp., Rhizoctonia solani J.G. Macrophomina phaseolina (Tassi) Goid. It is defined. Pre-patogenicity tests result from Alternaria spp. and Aspergillus niger isolat has not caused signs of brownness and dirt in the plant parts and therefore has not been taken to the trials of the seed. All the insulates of the macrophomina phaseolina and Rhizoctonia solani have been identified as pathogens in the crocodile plant, also supported by the tests carried out by trials.
