SüleymanDemirel Üniversitesi TIP FAKÜLTESİ DERGİSİ: 2004 Eylül; 11(3) Melatonin'in intestinal iskemi-reperfüzyonda 6-fosfoglukonat dehidrogenaz aktivitesi üzerine etkisi Burhan Ateş, H.Ramazan Yılmaz, Zeliha Selamoğlu, Mustafa Iraz ,İsmet Yılmaz Özet Bu çalışmada, intestinal iskemi-reperfüzyon (I/R) sürecinde, intestinal dokuda pentoz fosfat metabolik yolu- nun anahtar enzimlerinden biri olan 6-fosfoglukonat dehidrogenaz (6-PGD) enziminin aktivitesi ve melaton- in'in iskemi öncesi ve sonrası uygulanmasının enzim aktivitesi üzerine etkisi araştırıldı. 32 adet Sprague Dawley cinsi erkek sıçan, herbiri 8' er denekten oluşan 4 gruba ayrıldı (n: 8). Grup 1: plasebo, grup 2: I/R (Kontrol), grup 3: 10 mg/kg melatonin+I/R ve grup 4: I+10 mg/kg melatonin + R. I/R uygulaması, superior mezenterik arter (SMA)' in 45 dakika diseke edilmesi ve ardından 120 dakika reperfüzyonu ile gerçekleştiril- di. Tüm gruplarda enzim spesifik aktiviteleri, volüm aktivite değerleri total protein miktarlarına bölünerek hesaplandı. Elde edilen veriler, intestinal iskemi-reperfüzyon sürecinde 6-PGD aktivitesinde kontrol I/R grubunda plasebo grubuna kıyasla bir azalmanın olduğunu göstermekte, buna karşılık iskemi öncesi mela- tonin uygulanan grupta aktivite değerinin yükselerek plasebo grubuna yakın bir değer aldığı görülmektedir. Iskemiyi takiben melatonin verilen grupta ise enzim aktivitesi iskemi öncesi melatonin uygulanan gruptaki kadar bir artış göstermemiştir. Tüm gruplardaki farklılıklar istatistiksel olarak anlamlı değildir. Anahtar Sözcükler: Melatonin, Iskemi-reperfüzyon, 6-fosfoglukonat dehidrogenaz. Abstract The effect of melatonin on 6-phosphogluconate dehydrogenase in intestinal ischemia/reperfusion In this study, enzyme activity of 6-phosphogluconate dehydrogenase (6-PGD) which is one of the key enzyme in the intestinal tissue for intestinal ischemia/reperfusion (I/R) period in pentose phosphate metabolic path- way was investigated and the effect of melatonin on enzyme activity before and after I/R was determined. A total of 32 young male Sprague Dawley rats were divided into 4 groups (n= 8). Group 1 was placebo and Group 2 was control I/R group. Melatonin (10 mg/kg) and I/R was applied for Group 3 and melatonin (10g/kg) and R was applied for Group 4. I/R was performed by occlusion of superior mezenteric artery for 45 minutes and following reperfusion for 120 minutes. For all groups enzyme specific activity was calculated by dividing volume activity value to total protein amount. The results showed that there is a decrease in the activ- ity of pentose phosphate metabolic pathway key enzyme 6-phosphogluconate dehydrogenase (6-PGD) for control I/R group during intestinal ischemia- reperfusion period compare to sham group. But the activity val- ues of the group which melatonin was applied before ischemia was close to the values observed for then sham group. There was no obvious increase in the enzyme activity of the group which melatonin was applied after ischemia as compare to group which melatonin was applied before the ischemia.
