Elazığ’da Yetişen Polygonum cognatum Meissn (Madımak) Bitki Ekstraktlarının In vitro Biyolojik Aktiviteleri ve Bazı Fitokimyasal Bileşenlerinin Belirlenmesi

Halk arasında bitkilerin çeşitli kısımları kullanım amacına yönelik basit işlemlerden geçirilerek gerek gıda, gerekse tedavi amacıyla yaygın olarak kullanılmaktadır. Bitkiler, insan beslenmesinde çok önemli bir yer tutar ve insan sağlığı açısından bazı önemli bileşenler içermektedir. Dünyadaki bitkisel çeşitliliğin fazlalığı, çoğu bitkinin biyolojik etkileri ve etki mekanizmaları hakkındaki bilimsel verilerin hala yetersiz olması bu konuya olan ilgiyi her geçen gün artırmaktadır. Bu çalışmanın amacı, Elazığ yöresinden toplanan ve özellikle gıda olarak mevsiminde bolca tüketilen Polygonum cognatum Meissn (Madımak, Kuşekmeği) bitkisinin etanol, metanol, aseton ve hekzan ekstraktlarının antiradikal aktiviteleri, fitokimyasal bileşenleri, antimikrobiyal ve in vitro antikanserojen aktivitelerinin araştırılmasıdır. Çalışma sonuçlarımıza göre, P. cognatum Meissn özütlerinin ABTS radikal giderme aktivitelerinin DPPH a kıyasla daha iyi sonuç verdiği görüldü. Ayrıca ABTS radikal giderme aktivitelerinin standart bir antioksidan olan askorbik asite (Vitamin C) yakın değerlerde olduğu bulundu. Polygonum cognatum Meissn’ in yüksek miktarda fitosteroller, K ve D vitaminleri, α-tokoferol, flavonoidler, proantosiyanidin, fenolikler ve doymamış yağ asitleri içerdiği tespit edilmiştir. Bitki ekstraktlarının antimikrobiyal etkileri Escherichia coli, Bacillus subtilis, Klebsiella pneumoniae, Staphylococcus aureus and Candida albicans mikroorganizmaları kullanılarak Disk Difüzyon ve Minimum İnhibitör Konsantrasyonu (MİK) yöntemleri ile belirlendi. Bütün ekstraktların tüm mikroorganizmalara karşı antimikrobiyal aktivite gösterdiği görüldü. Aynı zamanda Polygonum cognatum Meissn ekstraktlarının, MCF-7, PC3 ve MKN-45 kanser hücre hatları üzerinde kontrole göre özellikle yüksek konsantrasyonlarda oldukça iyi sitotoksik etkiler sergilediği görüldü. Kanser hücreleri içerisinde en yüksek etkiyi ise MKN-45 (insan mide kanseri) hücre hattında göstermiştir. Elde edilen sonuçlar antioksidan ve antipiroliferatif etki gösteren ve biyoaktif bileşikleri içeren Polygonum cognatum Meissn bitkisinin insanlar tarafından tüketilmeye devam edilebileceğini göstermiştir.

Abstract:

The various parts of the plants among the people are used by simple processes for the purpose of use and are widely used for food or treatment purposes. Plants play a very important place in human nutrition and contain some important ingredients in terms of human health. The excess of plant diversity in the world, the lack of scientific data on the biological effects and mechanisms of most plants, increases interest in this issue every day. The purpose of this study is to investigate the anti-radical activities of the ethanol, methanol, acetone and hexane extract of the plant Polygonum cognatum Meissn, which is collected from the area of Elazığ and consumed especially as food in the season, and the phytochemical components, anti-microbial and in vitro anti-cancerogenic activities. According to our research results, P. cognatum Meissn extract shows that ABTS radical elimination activities have better results than DPPH. It was also found that ABTS radical removal activities were close to ascorbic acid (Vitamin C), a standard antioxidant. Polygonum cognatum Meissn has been found to contain high amounts of phytosterols, vitamins K and D, α-tokoferol, flavonoids, proantosyanidine, phenolics, and unsaturated fatty acids. Antimicrobial effects of plant extracts were determined by the use of microbials Escherichia coli, Bacillus subtilis, Klebsiella pneumoniae, Staphylococcus aureus and Candida albicans by the methods of Disk Diffusion and Minimum Inhibitor Concentration (MIC). It has been shown that all extracts have an antimicrobial activity against all microorganisms. It was also found that Polygonum cognatum Meissn extracts have quite good cytotoxic effects, especially at high concentrations, according to MCF-7, PC3 and MKN-45 cancer cell lines. The highest effect among cancer cells was shown in the cell line MKN-45 (human stomach cancer). The results found that the polygonum cognatum Meissn plant, which has an antioxidant and antipiroliferative effect and contains bioactive compounds, can continue to be consumed by humans.

The aim of this study, is to investigate the antiradical activities, phytochemical components, antimicrobial and in vitro anticancer activities of the ethanol, methanol, acetone and hexane extracts of the Polygonum cognatum Meissn (Madımak, Kuşekmeği) collected from Elazığ and consumed abundantly as a food in season. According to our study results, ABTS radical scavenging activities of Polygonum cognatum Meissn extracts were found to give better results compared to DPPH. Also, ABTS radical scavenging activities were found to be close to ascorbic acid, a standard antioxidant. It was established that Polygonum cognatum Meissn contains high amounts of phytosterols, vitamin K and D, α-tocopherol, flavonoids, proanthocyanidin, phenolics and unsaturated fatty acids. Antimicrobial effects of Polygonum cognatum Meissn extracts were determined by Disc Diffusion and the Minimum Inhibitory Concentration (MIC) methods using Escherichia coli, Bacillus subtilis, Klebsiella pneumoniae, Staphylococcus aureus and Candida albicans microorganisms. It was observed that all plant extracts showed antimicrobial activity against all microorganisms. However, Polygonum cognatum Meissn extracts were exhibited good cytotoxic effects on MCF-7, PC3 and MKN-45 cancer cell lines than control, especially at higher concentration. Among the cancer cells, MKN-45 (human stomach cancer) showed the highest effect in the cell lines. Consequently, obtained results indicate that Polygonum cognatum Meissn may continue to be consumed by humans owing to it has potentially antioxidant, antiproliferative properties and including bioactive compounds.
