Aktif Tüberkülozlu Türk Hastalarda Sitokin Sinyali Baskılayıcı Gen Polimorfizmlerinin Değerlendirilmesi

Amaç: Küresel bir sağlık sorunu olmaya devam eden tüberküloz (TB), çevresel faktörler ve konakçı genetik faktörlerinin etkisindedir. Bu çalışmada TB’li hastalarda sitokin sinyali baskılayıcı (SOCS)-1 geni promotör bölgesinde yer alan -1478CA/del (rs33989964) ve ekson-2 bölgesinde bulunan 1335G/C (rs11549428) tek nükleotid polimorfizmlerinin (single nücleotid polymorphism - SNP) TB’ye yatkınlık ya da direnç durumuyla ilişkisinin araştırılması amaçlanmıştır. Gereç ve Yöntem: Çalışmaya, klinik örneğinin kültüründe Mycobacterium tuberculosis kompleks izole edilen 90 TB hastası ile kontrol grubu olarak 90 sağlıklı kan donörü dahil edildi. Araştırılan SNP’lerin belirlenmesi için polimeraz zincir reaksiyonu ve restriksiyon parça uzunluk polimorfizm yöntemleri kullanıldı. Bulgular: SOCS-1 geni promotör bölgesinde bulunan -1478CA/del SNP’si, hasta ve sağlıklı kontrol grubunda Hardy-Weinberg dengesinde bulunurken, gruplar arasında allel sıklığı (p=0.327) ve genotip dağılımı (p=0.291) bakımından anlamlı bir ilişki tespit edilmedi. SOCS-1 geni ekson- 2’de bulunan 1335G/C SNP analizine göre hasta ve kontrol grubunun tamamında C alleli bulundu, istatistiksel değerlendirme yapılmadı. Sonuç: Çalışma sonucunda SOCS-1 geninde bulunan SNP’ler ile TB’ye direnç ya da duyarlılık arasında ilişki saptanmadı. Bu çalışma için araştırılması gereken SOCS-1 geni üstünde farklı polimorfizmlerin de olabileceği akılda tutulmalıdır.

Assessment of cytokine signal-pressive gen polymorphisms in active Turkish patients with tuberculosis

Abstract:

Goal: Tuberculosis (TB), which continues to be a global health problem, is influenced by environmental factors and host genetic factors. This study was aimed at investigating the relationship between single nucleotid polymorphisms (single nucleotid polymorphism - SNP) in patients with tuberculosis in the -1478CA/del (rs33989964) and 1335G/C (rs11549428) in the exon-2 in the cytokine signal suppressor (SOCS)-1 gene promoter area with the tendency or resistance to tuberculosis. The study included 90 healthy blood donors as a control group with 90 TB patients isolated from the Mycobacterium tuberculosis complex in the culture of the clinical sample. For the determination of the SNPs studied, polymerase chain reaction and restriction piece length polymorphism methods were used. Results: The -1478CA/del SNP in the SOCS-1 gene promoter area is in the Hardy-Weinberg balance in the patient and healthy control group, while there is no meaningful relationship between the groups in terms of allel frequency (p=0.327) and genotype distribution (p=0.291). According to the 1335G/C SNP analysis found in the SOCS-1 gene exon-2 all of the patient and control group C alelium, no statistical assessment was made. Result: The study found no relationship between the SNPs found in the SOCS-1 gene and resistance or sensitivity to TB. This study should be understood that different polymorphisms may also occur on the SOCS-1 gene that needs to be investigated.
