THE EFFECTS OF LARGE BOWEL PREPARATION TECHNIQUES ON THE MICROFLORA AND MUCOSAL STRUCTURE

We have investigated the effects of the large bowel preparation methods and antibiotics on the luminal and mucosal microflora of the colon. 35 rats (350-400 g Spraque Dawley) were divided into 7 groups. The first group was control. In the second group, 10% povidone iodine, in the third, fourth, fifth and sevenin groups magnesium Hydroxide and in the sixth group mannitol were given for the mechanical bowel preparation. Oral Ornidazole (20 mg/kg) for two days was added to the forth group. Oral Ornidazole (20 mg/kg) two days prior to the operation and parenteral Seftriakson (30 mg/kg) was given 1 hour before anesthesia to the fifth group. Oral Streptomycin (30 mg/kg) the day before surgery and Parenteral Ornidazol (30 mg/kg) 1 hour before anesthesia was given to the sixth group. Mezlocillin was given parenterally 1 hour before anesthesia to the seventh group. All rats were fed with a diet containing 50% meat for fifteen days to obtain close proximity of colon microflora to that of humans. Lumen contents and ceacum walls of all rats were cultured separately using standard qualitative and quantitative bacteriological techniques. Scanning electron microscopy was performed to determine the microflora in the colonic mucosa. Bacteriological results showed that mannitol, oral Streptomycin and parenteral Ornidazol group had the least bacterial colonisation in the colon wall and luminal contents.
