Amaç: Mastitisli ineklerin süt örneklerinden izole edilen ve fenotipik olarak metisilin dirençli Staphylococcus aureus (MRSA) olduğu belirlenen izolatlarda, mecC gen varlığının araştırılması amaçlandı. Gereç ve Yöntem: Çalışma materyalini mastitisli ineklerden izole edilen 150 adet Staphylococcus aureus (S. aureus) suşu oluşturdu. İzolatlardaki metisilin direnci sefoksitin disk difüzyon testi ve ticari PBP2a lateks aglütinasyon testi ile belirlendi. MRSA izolatlarında mecA ve mecC gen varlığı Polimeraz Zincir Reaksiyonu (PZR) ile analiz edildi. Bulgular: Yüz elli adet S. aureus izolatından 28’inin (%18.6) sefoksitin disk difüzyon testi ile fenotipik olarak metisilin dirençli olduğu belirlendi. MRSA izolatlarının 21’inde (%75) lateks test ile aglütinasyon ve PZR ile mecA geni belirlendi. Yedi (%25) izolatta aglütinasyon ve mecA geni belirlenemezken, mecC geni taşıdığı tespit edildi. Öneri: Türkiye’de mastitisli ineklerde mecC MRSA izolasyon oranının beklenmedik şekilde yüksek olduğu ve fenotipik olarak MRSA olarak identifiye edilen izolatların genotipik doğrulanmasında mecC geni yönünden de analiz edilmesi gerektiği kanaatine varıldı.
Purpose: In isolates that were isolated from dairy samples of Mastitis cows and phenotypically identified as methylococcus aureus (MRSA) resistant, the presence of mecC gene was intended to be investigated. Tools and Methods: The work material was formed by 150 Staphylococcus aureus (S. aureus), which was isolated from mastitised cows. The methysillin resistance in isolates was determined by the sefoxitin disk diffusion test and the commercial PBP2a latex aglutination test. The presence of mecA and mecC genes in MRSA isolates was analyzed with Polymerase Chain Reaction (PZR). The findings: 28 of a hundred and fifty S. aureus isolates (18.6%) were found to be phenotypically resistant to methysilin by the disc diffusion test. In 21 (75%) of MRSA isolates, the Latex test was identified with aglutination and PZR with mecA genes. Seven (25%) insulated aglutination and mecA genes were not identified, while mecC genes were found to be carried. Recommendation: In Turkey, the MCC MRSA isolation rate is unexpectedly high in mastitised cows and the phenotypically MRSA identified isolates should also be analyzed in the genotypical verification in the direction of the MCC gene.
