Introduction: Skin aging can be accelerated by UV exposure from the sun. UV exposure will increase oxidative stress to disturb collagen synthesis. To overcome the increase of oxidative stress, antioxidant compound from plants, such as coffee cascara pulp, can be utilized. Previous research proves that coffee cascara has the potential to overcome photoaging due to its content. This research aims to assess the benefits of topical cascara pulp arabica coffee extract cream on the number of collagen fibers thickness and density of Wistar rats (Rattus norvegicus) as a treatment for photoaging. Method: 30 male Wistar rats (Rattus norvegicus) was divided to 5 groups. The negative control group (PN) was given base cream and the positive control (PP) was given vitamin C 3%. The treatment group was given cascara pulp arabica coffee extract cream with 5% concentration (P1), 7.5 concentration (P2), and 10% concentration (P3). The group was exposed to 840 mJ/cm2 of UVB in total for 4 weeks. Skin biopsy was done to assess the thickness and density of collagen fiber through histological examination using masson trichrome staining. Result: The result shows significant differences (p<0.05) in collagen fiber thickness and density between the control and treatment groups. The topical application of cascara pulp arabica coffee extract cream gives higher collagen fiber density and thickness. Conclusion: Applying cascara pulp arabica coffee extract cream can protect and repair dermal collagen of male wistar rats (Rattus norvegicus) exposed to UVB.
