Koyun Dalak Doku Arjinazının Fotoinaktivasyonu ve Bazı Kinetik Özellikleri

Abstract:

Bu çalışmanın amacı koyun dalak doku arjinazının bazı kinetik özelliklerinin tesbit edilerek metilen mavisinin inhibisyon etkisinin araştırılmasıdır. Çalışmada kullanılan dalak dokuları Elazığ'daki Elkas kesimevinden temin edilmiştir. Arjinaz aktivitesi spektrofotometrik olarak tiyosemikarbazid-diasetilmonoksim üre (TDMU) metodu kullanılarak ölçüldü. Protein Lowry ve arkadaşlarının metoduna göre ölçülmüştür. Çalışmada dört grup kullanıldı. Birinci grupta, birinci preinkübasyonda MnCl2 eklendi ve örnekler oda ışığında tutuldu. Bu grup kontrol grubu olarak kabul edildi. İkinci grupta metilen mavisi ve MnCl2 birinci preinkübasyonda eklendi ve örnekler 150 watt ışığa maruz bırakıldı. Üçüncü grupta örneklere birinci preinkübasyonda MnCl2, ikici preinkübasyonda metilen mavisi eklenerek 150 w ışık uygulandı. Dördüncü grupta ise birinci preinkübasyonda metilen mavisi ikinci preinkübasyonda ise MnCl2 eklenerek örnekler 150 w ışığa tabi tutuldu. Koyun dalak doku arjinaz aktivitesinde en fazla kayıp dördüncü grupta olurken bunu ikinci grup takip etti. En az aktivite kaybı ise üçüncü grupta görüldü. Enzim aktivitesinde oda ışığında % 66, karanlıkta % 60 ve 150 w ışıkta % 72' lik bir inhibisyon olduğu saptandı. Metilen mavisinin koyun dalak doku arjinazı üzerinde non-kompetetif inhibisyona neden olduğu tesbit edildi. Fotoinaktivasyonun, muhtemelen arjinaz molekülünde bulunan histidil artıklarının imidazol gruplarının değişikliğe uğraması sonucu meydana geldiği düşünülmektedir.

Photoactivation of the Sheep Dalak Tissue Arginase and Some Kinetic Features

Abstract:

The purpose of this study is to investigate the inhibition effect of methylene blue by confirming some of the kinetic characteristics of the wheat tissue arginase. The wheels used in the study were provided from the Elkas cutting machine in Elazığ. Arginase activity was spectrophotometrically measured using the thyosemicarbazide-diacetylmonoxide urine (TDMU) method. Protein is measured by the method of Lowry and his friends. Four groups were used in the study. In the first group, MnCl2 was added in the first preincubation and the samples were kept in the room light. This group was recognized as a control group. In the second group, methylene blue and MnCl2 were added in the first preincubation, and the samples were exposed to 150 watt light. In the third group, the first preincubation MnCl2 was applied to the samples, adding methylene blue in the second preincubation 150 w light. In the fourth group, the first pre-incubation methyled blue in the second pre-incubation, and the MnCl2 added, the samples were subjected to 150 w light. The sheep's wing tissue was the largest loss in arginase activity in the fourth group, while the second group followed it. The lowest loss of activity was observed in the third group. In the enzyme activity, it was found to have a inhibition of 66% in room light, 60% in darkness and 72% in 150 w light. It has been found that methylene blue causes non-competitive inhibition on the dog's wave tissue arginase. Photoinactivation is believed to have occurred as a result of the changes in the imidazole groups of the hystidal remains found in the arjinase molecule.
