THE DEVELOPMENT OF CONDITIONS OF ISOLATION AND SPECTROPHOTOMETRIC DETERMINATION OF METFORMIN IN BIOLOGICAL OBJECTS

Abstract:

Objective. The development of conditions of metformin isolation from liver tissues in the directed chemical-toxicological analysis and its identification in obtained extracts by spectrophotometric method. Research methods. As research object substance of metformin hydrochloride has been applicated (monography EP 01/2005:0931, producer Harman Finochem Ltd.). Identification and quantitative determination were conducted using the modified spectrophotometer SF-46 LOMO. As model biological object sample grounded pork liver, saturated with aqueous solution of preparation, has been used. Results and discussion. The individual method of isolation metformin from liver tissues has been developed. Based on sufficiently high hydrophilic properties of metformin for its allotment from biological objects water has been proposed, and for its extraction from the aqueous phase into organic one mixture i-penthanol-buthanol (1:1) at pH 13 has been proposed. It has also been proposed conditions of purification of primary aqueous extracts of co-extractive acid substances with mixture hexane-toluene (1:1), and for purification of proteins has been proposed 25 % solution of sodium chloride. This method allows allocating toxicant from biological objects up to 82 %. Developing conditions of identification and quantitative determination of metformin in extracts from biological objects with spectrophotometric method the UV-specter of toxicant solution in methanol used, measured at the wavelength 236±2 nm. Content of metformin in extracts from biological objects has been determined by building the calibration graph. The relative standard deviation at quantitative determination of toxicant was 4,08 %, and the relative uncertainty of the average value was ± 11,36 %. Conclusions. Method of metformin isolation of liver tissues, applicable for goals of directed chemical-toxicological analysis, which allows allocating up to 82 % of preparation, has been developed. For its identification and quantitative determination in extracts from biological objects spectrophotometric method has been proposed.
