Amaç: Kalp, biyotoksik ajan olan Alüminyum (AL) birikimi için önemli bir hedef organdır. Bu çalışma, AL kaynaklı kardiyotoksisiteye karşı glutatyon öncüsü N-asetilsistein'nin (NAC), kardioprotektif etkisi olup olmadığını ve transient reseptör potansiyel melastatin 2 (TRPM2) kanalları ekspresyonu üzerine etkinliğini sıçan modelinde göstermeyi amaçlamaktadır. Gereç ve Yöntem: Sprague-Dawley cinsi erkek sıçanlar rastgele 4 eşit gruba ayrıldı (n=7). Kontrol grubuna herhangi bir uygulama yapılmadı. AL grubuna 14 gün boyunca 30 mg/kg/gün dozunda alüminyum klorür, intraperitonal (i.p.) olarak uygulandı. AL+NAC grubuna 14 gün boyunca 30 mg/kg/gün dozunda alüminyum klorür i.p. uygulandıktan 1 saat sonra 150 mg/kg/gün N-asetilsistein 14 gün boyunca i.p. olarak uygulandı. NAC grubuna ise 150 mg/kg/gün N-asetilsistein, 14 gün boyunca i.p. uygulandı. 14. günün sonunda sıçanlar dekapite edildi. Dekapitasyonun ardından kalp dokuları çıkarılarak biyokimyasal ve histolojik analizler yapıldı. Bulgular: AL grubunda histopatolojik değişiklikler ve miyokardiyal enzim seviyelerinde artış olduğu gözlemlendi. NAC uygulamasının AL kaynaklı histopatolojik değişiklikleri azalttığı gözlemlendi. Ayrıca AL grubunda Kontrol ve NAC gruplarına kıyasla TRPM2 ve Kaspaz 3 immünreaktivitesinde anlamlı bir artış olduğu belirlendi. AL+NAC grubunda ise AL grubuna göre TRPM2 ve Kaspaz 3 immünreaktivitesinde azalma olduğu belirlendi. Sonuç: AL kaynaklı kardiyotoksisite üzerine NAC uygulaması, biyokimyasal ve histopatolojik değişiklikleri azaltarak/hafifleterek ve TRPM2 kanalı ekspresyonunu düzenleyerek kardiyoprotektif etki gösterebilir.
The heart is an important target organ for the accumulation of aluminium (AL), a biotoxic agent. This study aims to show whether the glutation-pioneer N-acetilsysteine (NAC) against cardiotoxicity caused by AL has a cardioprotective effect and its effectiveness on the expression of the potential receptor melastatin 2 (TRPM2) channels in the mouse model. Method: Sprague-Dawley male rats were randomly divided into 4 equal groups (n=7). There was no application to the control group. Aluminum chloride in the dose of 30 mg/kg/day for the AL group for 14 days, intraperitonal (i.p.) as being applied. AL+NAC group for 14 days in 30 mg/kg/day dose of aluminum chloride i.p. 150 mg/kg/day N-acetilsysteine for 14 days after administration. as being applied. In the NAC group, 150 mg/kg/day of N-acetilsysteine for 14 days. was implemented. At the end of the 14th day, the mice was decaptivated. After the decapitation, the cardiovascular tissue was removed and biochemical and histological analyses were carried out. The findings: there were histopathological changes in the AL group and an increase in levels of myocardial enzymes. The application of NAC has been observed to reduce the histopathological changes caused by AL. It was also found that there was a significant increase in TRPM2 and Kaspaz 3 immune reactiveness in the AL group compared to the Control and NAC groups. In the AL+NAC group, there was a decrease in TRPM2 and Kaspaz 3 immune reactiveness according to the AL group. The application of NAC on cardiotoxicity caused by AL may have cardioprotective effects by reducing/reducing biochemical and histopathological changes and regulating TRPM2 channel expression.
