Amaç: Yara yeri infeksiyonunu engelleyerek iyi bir yara iyileşmesinin sağlanması önemli bir cerrahi sorundur. Geçmişten günümüze kadar pek çok çeşit bitkisel ve hayvansal ürünler birçok hastalık ve yaraların tedavisinde tıbbi amaçlarla kullanılagelmiştir. Bunlardan birisi de propolistir. NORs (Nucleolus Organizer Regions=Çekirdekçik Oluşturan Bölgeler) hücrenin canlılık, metabolik aktivite ve proliferasyonun iyi bir göstergesidir. Bu çalışmada, deneysel olarak oluşturulan cerrahi yara modelinde proliferasyon indeksinin bir göstergesi olarak da kullanılabilen NOR proteinlerinden nukleolin, nukleofosmin ve UBTF gen ekspresyon düzeyleri ve propolisin yara yeri infeksiyonunu önleme ve anastomoz iyileşmesine katkısını değerlendirerek literatüre katkıda bulunmayı amaçladık. Yöntem: Bu deneysel çalışma; cerrahi yaralar sınıflamasına göre temiz ve temiz-kontamine yaralar oluşturularak planlandı. Sıçanlar Kontrol, Anastomoz, Fusidik asit+Anastomoz ve Propolis+Anastomoz olarak dört ana gruba ayrıldı. Toplam 4 hayvan grubu, her bir grupta sekizer adet ve toplamda 32 hayvan kullanıldı. Hayvanlarda yara yeri infeksiyon belirteç olarak hemogram ve anastomoz dokusunda NCL, NPM1 ve UBTF gen ekspresyon düzeyleri çalışılarak karşılaştırıldı. Bulgular: Tüm gruplar dikkate alındığında, NCL (p=0,038), NPM1 (p=0,001) ve UBTF (p=0.000) gen ekspresyon düzeyleri için gruplar arasında istatistiksel olarak anlamlı farklılık saptandı. Tüm gruplar dikkate alındığında, cerrahi yara infeksiyonuna bağlı olarak, kan parametreleri açısından gruplar arasında istatistiksel olarak anlamlı farklılıklar tespit belirlendi (p<0.05). Sonuç: NCL, NPM1 ve UBTF gibi nükleolar proteinler; hücre homeostazının sürdürülmesi için önemli işlevlere sahiptir ve cerrahi yaralarda infeksiyonu engelleyerek anastomoz iyileşmesinin sağlanması için belirteç olarak kullanılabilir.

Purpose: Providing a good wound healing by preventing the wound site infection is an important surgical problem. From the past to today, many kinds of plant and animal products have been used for medical purposes in the treatment of many diseases and wounds. One of them is propolis. NORs (Nucleolus Organizer Regions) are a good indicator of cell vitality, metabolic activity and proliferation. In this study, we aimed to contribute to literature by evaluating the levels of nucleolin, nucleofosmin and UBTF gen expression and the contribution of propolis to the prevention of wound site infection and the healing of anastomosis from NOR proteins, which can also be used as an indicator of the proliferation index in the experimentally created wound model. Method: This experimental study was planned by creating clean and clean-contaminant wounds according to the classification of surgical wounds. The rats were divided into four main groups: Control, Anastomose, Fusic Acid+Anastomose and Propolis+Anastomose. A total of 4 groups of animals, eight hundred in each group and a total of 32 animals were used. In animals the wound place infection indicator was compared by studies of the levels of expression of the NCL, NPM1 and UBTF genes in the hemogram and anastomosus tissue. Results: When all groups were taken into account, statistically significant differences were found between the groups for NCL (p=0,038), NPM1 (p=0,001) and UBTF (p=0.000) genetic expression levels. When all groups were taken into account, statistically significant differences between the groups were identified in terms of blood parameters, depending on the surgical injury infection (p<0.05). Nuclear proteins such as NCL, NPM1 and UBTF have important functions for the maintenance of cell homeostasis and can be used as a indicator for the guarantee of anastomosis recovery by preventing infection in the wounds.

We aimed to detect Nucleolin, Nucleophosmin and Upstream Binding Transcription Factor gene expressions from NOR proteins, which can also be used as an indicator of proliferation index in the experimentally surgical wound model and contribution of propolis to anastomotic healing by preventing wound infection, thus we can contribute to the literature. Method: This experimental study was planned by creating clean and clean-contaminated wounds according to the surgical wounds classification. Rats were divided into four main groups as Control, Anastomosis, Fusidic acid + Anastomosis and Propolis + Anastomosis. A total of 4 groups of animals, eight in each group, and a total of 32 animals were used. NCL, NPM1 and UBTF gene expression levels at anastomosis tissue and hemogram were studied and compared in as the markers of wound infection in animals. Results: It was observed that there were statistically significant differences between the all groups in terms of NCL (p=0.038), NPM1 (p=0.001) and UBTF (p=0.001) gene expression levels and hemogram parameters. Considering all groups, a statistically significant differences were found between the groups in terms of blood parameters, depending on surgical wound infection (p <0.05). Conclusion: Nucleolar proteins such as NCL, NPM1 and UBTF have important functions in maintaining cell homeostasis and can be used as a marker to prevent infection in surgical wounds and to ensure anastomotic healing.
